Abstract

The disclosure provides novel molecules related to growth and differentiation factor-8 (GDF-8), in particular mouse and humanized antibodies, and antibody fragments, including those that inhibit GDF-8 activity and signaling in vitro and/or in vivo. The disclosure also provides methods for diagnosing, treating, ameliorating, preventing, prognosing, or monitoring degenerative orders of muscle, bone, and insulin metabolism, etc., in particular amyotrophic lateral sclerosis (ALS). In addition, the disclosure provides pharmaceutical compositions for the treatment of such disorders by using the antibodies, polypeptides, polynucleotides, and vectors of the invention.

Claims (29)

1. An isolated antibody that binds GDF-8, comprising a heavy chain variable region comprising three complementarity determining regions (CDRs) comprising the amino acid sequences of SEQ ID NO: 10, SEQ ID NO: 11, and SEQ ID NO: 12, and a light chain variable region comprising three CDRs comprising the amino acid sequences of SEQ ID NO: 13, SEQ ID NO: 14, and SEQ ID NO: 15.

2. An isolated antibody that binds GDF-8, comprising a heavy chain variable region wherein said heavy chain variable region comprises a CDR1 comprising the amino acid sequence of SEQ ID NO: 10, a CDR2 comprising the amino acid sequence of SEQ ID NO: 11, and a CDR3 comprising the amino acid sequence of SEQ ID NO: 12, and a light chain variable region wherein said light chain variable region comprises a CDR1 comprising the amino acid sequence of SEQ ID NO: 13, a CDR2 comprising the amino acid sequence of SEQ ID NO: 14, and a CDR3 comprising the amino acid sequence of SEQ ID NO: 15.

3. The antibody of claim 2 , wherein the antibody comprises a heavy chain variable region comprising an amino acid sequence selected from the group consisting of

the amino acid sequence of SEQ ID NO: 3, and the amino acid sequence of SEQ ID NO:7,

and a light chain variable region comprising an amino acid sequence selected from the group consisting of

the amino acid sequence of SEQ ID NO:5, and the amino acid sequence of SEQ ID NO; 9.

4. The antibody of claim 3 , wherein the antibody is selected from the group consisting of:

a) an antibody comprising a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:7, and a light chain variable region comprising the amino acid sequence of SEQ ID NO:9;

b) an antibody comprising a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:3, and a light chain variable region comprising the amino acid sequence of SEQ ID NO:5;

c) an antibody comprising a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:7, and a light chain variable region comprising the amino acid sequence of SEQ ID NO:5; and

d) an antibody comprising a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:3, and a light chain variable region comprising the amino acid sequence of SEQ ID NO:9.

5. The antibody of claim 1 , wherein said antibody comprises a heavy chain variable region comprising the amino acid sequence of SEQ ID NO:7, and a light chain variable region comprising the amino acid sequence of SEQ ID NO:9.

6. An isolated antibody capable of binding GDF-8, comprising the amino acid sequences of SEQ ID NO: 10, SEQ ID NO: 11, SEQ ID NO: 12, SEQ ID NO: 13, SEQ ID NO: 14, and SEQ ID NO: 15.

7. An isolated antibody capable of binding GDF-8 comprising a heavy chain variable region comprising the amino acid sequence encoded by the DNA sequence of SEQ ID NO: 2 (VH), and a light chain variable region comprising the amino acid sequence encoded by the DNA sequence of SEQ ID NO: 4 (LH).

8. An isolated antibody capable of binding GDF-8 comprising a heavy chain variable region comprising the amino acid sequence encoded by the DNA sequence of SEQ ID NO: 6 (VH), and a light chain variable region comprising the amino acid sequence encoded by the DNA sequence of SEQ ID NO: 8 (LH).

9. An isolated antibody capable of binding GDF-8 said antibody comprising the amino acid sequences of SEQ ID NO:7 (VH) and SEQ ID NO:9 (VL).

10. An isolated antibody capable of binding GDF-8 wherein the antibody comprises: the amino acid sequence of SEQ ID NO:3 and the amino acid sequence of SEQ ID NO:5.

11. The isolated antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , further comprising a heavy chain constant region chosen from IgG1, IgG2, IgG3, and IgG4.

12. The isolated antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , further comprising a light chain constant region chosen from kappa or lambda.

13. The isolated antibody of claim 11 , wherein the constant region comprises the amino acid sequence of SEQ ID NO:19.

14. The isolated antibody of claim 13 , wherein the amino acids of SEQ ID NO:19 are mutated at residues 117 and 120.

15. The antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , wherein the antibody binds to GDF-8 with a Kd of at least 10 −7 M, 10 −8 M, 10 −10 M or 10 −11 M.

The isolated antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , wherein the antibody is selected from the group consisting of an intact antibody, an Fab fragment, an F(ab′) 2 fragment, a scFV fragment, a monoclonal antibody, a humanized monoclonal antibody, a chimeric antibody, and an Fv fragment.

17. The antibody of claim 16 , wherein the antibody is a monoclonal antibody.

18. The antibody of claim 16 , wherein the antibody is a humanized monoclonal antibody.

19. The isolated antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , wherein the glycosylation pattern of the antibody is modified.

20. The isolated antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 , wherein the antibody is labeled.

21. An isolated antibody produced by a method comprising culturing a host cell comprising a polynucleotide encoding an antibody that binds to GDF-8, wherein the polynucleotide comprises DNA sequences selected from the group consisting of a) SEQ ID NO:2 and SEQ ID NO:4, and b) SEQ ID NO:6 and SEQ ID NO:8, expressing the antibody and recovering the antibody expressed by the host cell.

22. A pharmaceutical composition comprising a pharmaceutically acceptable carrier and at least one antibody selected from the group consisting of the antibody of any of claims 1 , 2 , 3 - 5 , 6 , 7 , 8 , 9 and 10 .
