Nucleic acid encoding a self-assembling split-fluorescent protein system
View Patent ↗The invention provides a protein labeling and detection system based on self-complementing fragments of fluorescent and chromophoric proteins. The system of the invention is exemplified with various combinations of self-complementing fragments derived from Aequorea victoria Green Fluorescent Protein (GFP), which are used to detect and quantify protein solubility in multiple assay formats, both in vitro and in vivo.
1. An isolated nucleic acid molecule comprising a polynucleotide encoding an 11 beta-strand polypeptide of a fluorescent protein, wherein the 11 beta-strand polypeptide has an amino acid sequence selected from the group consisting of SEQ ID NOS: 12, 14, 16, 18 and 20, and wherein the 11 beta-strand polypeptide is capable of self-complementation with a polypeptide corresponding to beta-strands 1-10 of the fluorescent protein.
2. The isolated nucleic acid molecule of claim 1 , wherein the polynucleotide has the nucleotide sequence of SEQ ID NO: 11.
3. The isolated nucleic acid molecule of claim 1 , wherein the polynucleotide has the nucleotide sequence of SEQ ID NO: 13.
4. The isolated nucleic acid molecule of claim 1 , wherein the polynucleotide has the nucleotide sequence of SEQ ID NO: 15.
5. The isolated nucleic acid molecule of claim 1 , wherein the polynucleotide has the sequence of SEQ ID NO: 17.
6. The isolated nucleic acid molecule of claim 1 , wherein the polynucleotide has the nucleotide sequence of SEQ ID NO: 19.
7. An expression vector comprising the nucleic acid molecule of any one of claims 1 and 2 - 6 .
8. A host cell transformed with the expression vector of claim 7 .
9. An isolated polynucleotide encoding an 11 beta-strand polypeptide of a Green Fluorescent Protein (GFP), wherein the 11 beta-strand polypeptide has the amino acid sequence of SEQ ID NO: 12, and wherein the 11 beta-strand polypeptide is capable of self-complementation with a polypeptide corresponding to beta-strands 1-10 of said GFP.
