Abstract

The invention provides a non-naturally occurring microbial organism having an adipate, 6-aminocaproic acid or caprolactam pathway. The microbial organism contains at least one exogenous nucleic acid encoding an enzyme in the respective adipate, 6-aminocaproic acid or caprolactam pathway. The invention additionally provides a method for producing adipate, 6-aminocaproic acid or caprolactam. The method can include culturing an adipate, 6-aminocaproic acid or caprolactam producing microbial organism, where the microbial organism expresses at least one exogenous nucleic acid encoding an adipate, 6-aminocaproic acid or caprolactam pathway enzyme in a sufficient amount to produce the respective product, under conditions and for a sufficient period of time to produce adipate, 6-aminocaproic acid or caprolactam.

Claims (16)

1. A host microbial organism in which exogenous nucleic acids are introduced, wherein said exogenous nucleic acids encode adipate pathway enzymes expressed in a sufficient amount to produce adipate, said adipate pathway comprising a succinyl-CoA:acetyl-CoA acyl transferase; a 3-oxoadipyl-CoA transferase; a 3-oxoadipate reductase; a 3-hydroxyadipate dehydratase; and a 2-enoate reductase.

2. A method for producing adipate, comprising culturing the host microbial organism of claim 1 under conditions and for a sufficient period of time to produce adipate.

3. The method of claim 2 , wherein at least one exogenous nucleic acid is a heterologous nucleic acid.

4. The method of claim 2 , wherein said host microbial organism is in a substantially anaerobic culture medium.

5. The host microbial organism of claim 1 , wherein host microbial organism comprises an adipyl-CoA pathway, wherein said adipyl-CoA pathway converts adipate to adipyl-CoA.

The host microbial organism of claim 5 , wherein said adipyl-CoA pathway comprises an exogenous nucleic acid encoding an adipyl-CoA pathway enzyme selected from an adipyl-CoA synthetase, a phosphotransadipylase, an adipate kinase, an adipyl-CoA:acetyl-CoA transferase and an adipyl-CoA hydrolase.

7. The host microbial organism of claim 6 , wherein said adipyl-CoA pathway comprises an exogenous nucleic acid encoding an adipyl-CoA synthetase.

8. The host microbial organism of claim 6 , wherein said adipyl-CoA pathway comprises an exogenous nucleic acid encoding a phosphotransadipylase and an adipate kinase.

9. The host microbial organism of claim 6 , wherein said adipyl-CoA pathway comprises an exogenous nucleic acid encoding an adipyl-CoA:acetyl-CoA transferase.

10. The host microbial organism of claim 6 , wherein said adipyl-CoA pathway comprises an exogenous nucleic acid encoding an adipyl-CoA hydrolase.

The host microbial organism of claim 5 , wherein said host microbial organism further comprises a 6-aminocaproic acid pathway, wherein said 6-aminocaproic acid pathway comprises exogenous nucleic acids encoding 6-aminocaproic acid pathway enzymes expressed in a sufficient amount to produce 6-aminocaproic acid, said 6-aminocaproic acid pathway enzymes comprising a CoA-dependent aldehyde dehydrogenase and a 6-aminocaproate dehydrogenase; or a CoA-dependent aldehyde dehydrogenase and a transaminase, wherein said transaminase converts adipate semialdehyde to 6-aminocaproic acid.

12. A method for producing 6-aminocaproic acid, comprising culturing the host microbial organism of claim 11 under conditions and for a sufficient period of time to produce 6-aminocaproic acid.

13. The method of claim 12 , wherein at least one exogenous nucleic acid is a heterologous nucleic acid.

14. The method of claim 12 , wherein said host microbial organism is in a substantially anaerobic culture medium.

15. The host microbial organism of claim 1 , wherein at least one exogenous nucleic acid is a heterologous nucleic acid.

16. The host microbial organism of claim 1 , wherein said host microbial organism is in a substantially anaerobic culture medium.
