Abstract

The invention relates to the identification and generation of antibodies that bind to TMEFF2 proteins which are involved in cancer. Further, the invention provides for the use of these antibodies in the treatment of cancer.

Claims (33)

1. An isolated antibody or antigen binding fragment that binds a TMEFF2 protein and competes for binding to said protein with TMEFF2#19 antibody, wherein the TMEFF2#19 antibody is produced by the hybridoma cell line deposited as ATCC Accession No: PTA-4127 and wherein the antibody or binding fragment comprises the complementary determining regions (CDRs) of SEQ ID NO: 28 which are present in a heavy chain variable region, and the CDRs of SEQ ID NO: 32 which are present in a light chain variable region.

2. The antibody of claim 1 , wherein the TMEFF2 protein is on a prostate cancer cell.

3. The antibody of claim 2 , wherein the prostate cancer cell is in a patient.

4. The antibody of claim 1 , wherein the antigen binding fragment is a Fab or F(ab) 2 fragment.

5. The antibody of claim 1 , wherein the antibody is humanized.

6. The antibody of claim 1 , wherein the antibody is a monoclonal.

7. The antibody of claim 1 , wherein the antibody is conjugated to a detectable label.

8. The antibody of claim 7 , wherein the detectable label is a fluorescent label.

9. The antibody of claim 1 , wherein the antibody is conjugated to an effector component selected from the group consisting of a toxin, a chemotherapeutic agent, and a radioisotope.

10. The antibody of claim 9 , wherein the toxin is selected from the group consisting of diphtheria A, exotoxin A, ricin A, abrin A, curcin, crotin, phenomycin, neomycin, and auristatin.

11. The antibody of claim 10 , wherein the toxin is auristatin.

12. The antibody of claim 1 comprising the heavy chain variable region selected from the group consisting of an amino acid sequence of SEQ ID NOs: 22, 24, and 28; and the light chain variable region selected from the group consisting of an amino acid sequence of SEQ ID NOs: 30 and 32.

13. The antibody of claim 12 , wherein:

the heavy chain variable region comprises the amino acids of SEQ ID NO: 22 and the light chain variable region comprises the amino acids of SEQ ID NO: 30;

the heavy chain variable region comprises the amino acids of SEQ ID NO: 22 and the light chain variable region comprises the amino acids of SEQ ID NO: 32;

the heavy chain variable region comprises the amino acids of SEQ ID NO: 24 and the light chain variable region comprises the amino acids of SEQ ID NO: 30;

the heavy chain variable region comprises the amino acids of SEQ ID NO: 24 and the light chain variable region comprises the amino acids of SEQ ID NO: 32;

the heavy chain variable region comprises the amino acids of SEQ ID NO: 28 and the light chain variable region comprises the amino acids of SEQ ID NO: 30; or

the heavy chain variable region comprises the amino acids of SEQ ID NO: 28 and the light chain variable region comprises the amino acids of SEQ ID NO: 32.

The antibody of claim 1 , wherein the antibody comprises a protein comprising the heavy chain variable region encoded by a nucleic acid sequence selected from the group consisting of SEQ ID NOs: 21, 23, and 27; and the light chain variable region encoded by a nucleic acid sequence selected from the group consisting of SEQ ID NOs: 29 and 31.

15. The antibody of claim 14 , wherein:

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 21 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 29;

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 21 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 31;

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 23 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 29;

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 23 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 31;

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 27 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 29; or

the heavy chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 27 and the light chain variable region is encoded by the nucleic acid sequence of SEQ ID NO: 31.

16. The antibody of claim 14 , wherein the antibody is conjugated to a detectable label.

17. The antibody of claim 16 , wherein the detectable label is a fluorescent label.

18. The antibody of claim 14 , wherein the antibody is conjugated to an effector component selected from the group consisting of a toxin, a chemotherapeutic agent, and a radioisotope.

19. The antibody of claim 18 , wherein the toxin is selected from the group consisting of diphtheria A, exotoxin A, ricin A, abrin A, curcin, crotin, phenomycin, neomycin, and auristatin.

20. The antibody of claim 19 , wherein the toxin is auristatin.

21. A pharmaceutical composition comprising a pharmaceutically acceptable excipient and the antibody of any one of claims 1 - 20 .
