Abstract

Disclosed are methods of identifying an agent that modulates an NFAT regulator protein. One such method comprises contacting at least one test agent with a recombinant cell comprising at least one NFAT regulator protein or fragment or derivative thereof, assessing the effect of the test agent on an activity, interaction, expression, or binding to the NFAT regulator protein or fragment or derivative thereof, and identifying the test agent that has an effect on an activity, interaction, expression, or binding to the NFAT regulator protein or fragment or derivative thereof, whereby the identified test agent is characterized as an agent that modulates an NFAT regulator protein. Methods of identifying an agent that modulates intracellular calcium, methods to screen for an agent that modulates NFAT regulator function, methods to diagnose unexplained immunodeficiency in a subject, and methods for identifying an agent for treating or preventing a disease or disorder associated with a NFAT regulator protein or calcium signaling are also disclosed.

Claims (13)

1. An isolated recombinant cell comprising heterologous nucleic acid encoding at least one Nuclear Factor of Activated T cells (NFAT) regulator protein, wherein the heterologous nucleic acid is a mammalian NFAT regulator gene, wherein the NFAT regulator gene is ORAI1, ORAI2, ORAI3, wherein ORAI 1 is encoded by a nucleic acid sequence comprising SEQ ID NO: 1 or a sequence 95% homologous thereto, ORAI 2 is encoded by a nucleic acid sequence comprising SEQ ID NO:2 or a sequence 95% homologous thereto, ORAI 3 is encoded by a nucleic acid sequence comprising SEQ ID NO:3 or a sequence 95% homologous thereto, and wherein the cell comprises a heterologous calcium modulating agent.

2. The recombinant cell of claim 1 , wherein the cell is a mammalian, human, rodent, insect or Xenopus cell.

3. The recombinant cell of claim 1 , further comprising a heterologous mammalian NFAT regulator gene, wherein the NFAT regulator gene is stromal interaction molecule (STIM) gene.

4. The recombinant cell of claim 3 , wherein the heterologous nucleic acid encoding ORAI and STIM are expressed in the cell to produce heterologous polypeptides.

5. The recombinant cell of claim 1 , wherein the NFAT regulator gene is encoded by SEQ ID: No:1, SEQ ID No:2, or SEQ ID No:3.

6. An in vitro method for screening for a test agent that modulates the activity of an ORAI polypeptide that is ORAI 1, ORAI 2 or ORAI 3, the method comprising:

a) providing a cell, wherein the cell expresses a recombinant mammalian ORAI polypeptide encoded by a nucleic acid sequence comprising any one of the nucleic acid sequence selected from the group consisting of SEQ ID No. 1, SEQ ID No. 2, SEQ ID No. 3, a sequence 95% homologous to SEQ ID No. 1, a sequence 95% homologous to SEQ ID No. 2, and a sequence 95% homologous to SEQ ID No. 3 to form a calcium release-activated Ca 2+ (CRAC) channel;

b) contacting the cell with a test agent; and

c) measuring store-operated calcium entry of the calcium release-activated Ca2+ (CRAC) channel,

wherein a change in the store-operated calcium entry of the calcium release-activated Ca2+ (CRAC) channel as compared to store-operated calcium entry of the calcium release-activated Ca2+ (CRAC) channel in the absence of the test agent indicates that the test agent is capable of modulating the activity of the calcium release-activated Ca2+ (CRAC) channel.

7. The method of claim 6 wherein the ORAI polypeptide is an ORAI1 polypeptide encoded by SEQ ID NO:1 or a sequence with 95% homology thereto.

8. The method of claim 6 wherein the ORAI polypeptide is an ORAI2 polypeptide encoded by SEQ ID NO:2 or a sequence with 95% homology thereto.

9. The method of claim 6 wherein the ORAI polypeptide is an ORAI3 polypeptide encoded by SEQ ID NO:3 or a sequence with 95% homology thereto.
