Use of pleitrophin for preventing and treating pancreatic diseases and/or obesity and/or metabolic syndrome
The present invention discloses the protein pleiotrophin secreted by the developing pancreas, and polynucleotides, which identify and encode this protein. The invention also relates to the use of these sequences in the diagnosis, study, prevention, and treatment of pancreatic diseases (e.g. diabetes), obesity, and/or metabolic syndrome.
1. A method for differentiating stem cells into insulin-producing cells comprising culturing said stem cells in a culture medium comprising human pleiotrophin (SEQ ID NO: 2) or a fragment thereof in an amount effective to produce insulin-producing cells.
2. The method according to claim 1 , wherein the stem cells have been modified to constitutively express the PAX-4 gene.
3. The method according to claim 1 , wherein the stem cells are embryonic stem cells, adult stem cells, somatic stem cells or primordial germ cells.
4. The method according to claim 1 , wherein the stem cells are of human origin.
5. The method according to claim 1 , wherein the stem cells are cultured with human pleiotrophin comprising SEQ ID NO: 2.
6. The method according to claim 1 , wherein the stem cells are aggregated to form embryoid bodies.
