Abstract

Anti-CLL-1 antibodies and antigen-binding fragments thereof, as well as pharmaceutical compositions comprising such antibodies and antigen-binding fragments are described. Also described are methods of using such antibodies and antigen-binding regions to bind CLL-1 and treat diseases, such as hematologic malignancies, which are characterized by expression of CLL-1.

Claims (16)

1. An isolated antibody or antigen-binding fragment thereof, comprising:

a) a heavy chain comprising a first variable region comprising CDR1 (amino acids 49 to 58), CDR2 (amino acids 73 to 89), and CDR3 (amino acids 122 to 132) from SEQ ID NO: 5; and

b) a light chain comprising a second variable region comprising CDR1 (amino acids 47 to 56), CDR2 (amino acids 72 to 78), and CDR3 (amino acids 111 to 120) from SEQ ID NO: 7,

wherein the antibody binds CLL-1 with high affinity.

2. The isolated antibody or antigen-binding fragment thereof of claim 1 , wherein:

a) the heavy chain, comprising a first variable region, comprises a sequence that has at least 99% identity to the amino acid sequence as set forth in SEQ ID NO: 5; and

b) the light chain, comprising a second variable region, comprises a sequence that has at least 99% identity to the amino acid sequence as set forth in SEQ ID NO: 7,

wherein the antibody binds to CLL-1 with high affinity.

3. The antibody of claim 1 , wherein the heavy chain and the light chain are connected by a flexible linker to form a single chain antibody.

4. The antibody of claim 3 , which is a single-Fv antibody.

5. The antibody of claim 1 , which is selected from the group consisting of an Fab antibody, an Fab′ antibody, an (Fab′) 2 antibody, a humanized antibody, and a chimeric antibody.

6. A pharmaceutical composition comprising the antibody of claim 1 , further comprising a suitable carrier.

7. An isolated cell line that produces an antibody according to claim 1 .

8. The antibody of claim 1 , wherein said antibody is a humanized form thereof.

9. The antibody of claim 1 , wherein said antibody is conjugated to a cytotoxic molecule.

10. The antibody of claim 9 , wherein said cytotoxic molecule is selected from the group consisting of a radioisotope, a toxin, a chemotherapeutic agent, a drug, and a growth inhibitory agent.
