Peptide-based in vivo siRNA delivery system
The present invention is directed compositions for targeted delivery of RNA interference (RNAi) polynucleotides to hepatocytes in vivo. Targeted RNAi polynucleotides are administered together with co-targeted melittin delivery peptides. Delivery peptides provide membrane penetration function for movement of the RNAi polynucleotides from outside the cell to inside the cell. Reversible modification provides physiological responsiveness to the delivery peptides.
1. A reversibly masked melittin peptide having the structure represented by
Melittin-(L-T) x
wherein Melittin is a melittin peptide, L is a physiologically labile reversible linkage, T comprises a targeting ligand, and x is an integer greater than zero.
2. A composition comprising a plurality of the reversibly masked melittin peptides of claim 1 , wherein x is an integer having a value greater than 80% of the number of primary amines of the plurality of melittin peptides.
3. The composition of claim 2 , wherein x is an integer having a value greater than 90% of the number of primary amines of the plurality of melittin peptides.
4. The reversibly masked melittin peptide of claim 1 wherein the Melittin peptide is selected from the list consisting of: Seq. ID 1, Seq. ID 7, Seq. ID 11, Seq. ID 51, Seq. ID 57, Seq. ID 58, Seq. ID 92, and Seq. ID 96.
5. The reversibly masked melittin peptide of claim 1 wherein the Melittin peptide consists of D-form amino acids.
6. The reversibly masked melittin peptide of claim 1 wherein the physiologically labile reversible linkage is a disubstituted maleamate.

8. The reversibly masked melittin peptide of claim 1 wherein the physiologically labile reversible linkage is a dipeptide-amidobenzyl carbamate.
9. The reversibly masked melittin peptide of claim 6 wherein-(L-T) has the structure represented by:
wherein R4 comprises the targeting ligand, R2 comprises a hydrophobic amino acid side group and R1 comprises an neutral amino acid side group.
10. The reversibly masked melittin peptide of claim 1 further comprising a polyethyleneglycol (PEG) covalently linked to the amino terminus of the melittin peptide.
11. The reversibly masked melittin peptide of claim 1 further comprising a targeting ligand- PEG conjugate covalently linked to the amino terminus of the melittin peptide.
12. The reversibly masked melittin peptide of claim 1 wherein an RNA interference polynucleotide is linked to the melittin peptide.
13. The reversibly masked melittin peptide of claim 1 wherein the targeting ligand comprises an Asialoglycoprotein Receptor (ASGPr) ligand.
14. The reversibly masked melittin peptide of claim 13 wherein the ASGPr ligand is selected from the group consisting of lactose, galactose, N-acetylgalactosamine (GalNAc), galactosamine, N-formylgalactosamine, N-propionylgalactosamine, N-n-butanoyl-galactosamine, and N-iso-butanoyl-galactosamine.
