Abstract

A method and device for treating a mammalian organism to obtain a desired local or systemic physiological or pharmacological effect is provided. The method includes administering a sustained release drug delivery system to a mammalian organism in need of such treatment at an area wherein release of an effective agent is desired and allowing the effective agent to pass through the device in a controlled manner. The device includes an inner core or reservoir including the effective agent, an impermeable tube which encloses portions of the reservoir, and a permeable member at an end of the tube.

Claims (20)

1. A sustained release drug delivery system suitable for injection into the vitreous of the eye comprising:

a drug reservoir comprising a therapeutically effective amount of an agent;

an inner tube having first and second open ends, and covering at least a portion of said drug reservoir, wherein said inner tube is permeable to the passage of the agent, dimensionally stable, and capable of supporting its own weight; and

an outer layer, permeable to the passage of said agent, covering only a portion of said inner tube, wherein:

the diameter of the tube is not more than 3 millimeters; and

upon implantation, agent is released through at least one of the open ends.

2. A sustained release drug delivery system in accordance with claim 1 , further comprising an impermeable member positioned at said inner tube first open end or said inner tube second open end.

3. A sustained release drug delivery system in accordance with claim 1 , further comprising a permeable member positioned at said inner tube first open end and/or said inner tube second open end.

4. A sustained release drug delivery system in accordance with claim 1 , further comprising a permeable member positioned at said inner tube first open end and an impermeable member positioned at said inner tube open second end.

5. A sustained release drug delivery system in accordance with claim 1 , further comprising an impermeable member positioned at said inner tube first open end and a permeable member positioned at said inner tube open second end.

A sustained release drug delivery system in accordance with claim 1 , wherein the inner tube is formed of a material selected from ethyl vinyl acetate, polyvinyl acetate, cross-linked polyvinyl alcohol, cross-linked polyvinyl butyrate, ethylene ethylacrylate copolymer, polyethyl hexylacrylate, polyvinyl chloride, polyvinyl acetals, plasticized ethylene vinylacetate copolymer, polyvinyl alcohol, polyvinyl acetate, ethylene vinylchloride copolymer, polyvinyl esters, polyvinylbutyrate, polyvinylformal, polyamides, polymethylmethacrylate, polybutyl methacrylate, plasticized polyvinyl chloride, plasticized nylon, plasticized soft nylon, plasticized polyethylene terephthalate, natural rubber, polyisoprene, polyisobutylene, polybutadiene, polyethylene, polytetrafluoroethylene, polyvinylidene chloride, polyacrylonitrile, cross-linked polyvinylpyrrolidone, polytrifluorochloroethylene, chlorinated polyethylene, poly(1,4′-isopropylidene diphenylene carbonate), vinylidene chloride, acrylonitrile copolymer, vinyl chloride-diethyl fumarate copolymer, silicone rubbers, ethylene-propylene rubber, silicone-carbonate copolymers, vinylidene chloride-vinyl chloride copolymer, vinyl chloride-acrylonitrile copolymer, and vinylidene chloride-acrylonitrile copolymer.

A sustained release drug delivery system in accordance with claim 1 , wherein the outer layer is formed of a material selected from ethyl vinyl acetate, polyvinyl acetate, cross-linked polyvinyl alcohol, cross-linked polyvinyl butyrate, ethylene ethylacrylate copolymer, polyethyl hexylacrylate, polyvinyl chloride, polyvinyl acetals, plasticized ethylene vinylacetate copolymer, polyvinyl alcohol, polyvinyl acetate, ethylene vinylchloride copolymer, polyvinyl esters, polyvinylbutyrate, polyvinylformal, polyamides, polymethylmethacrylate, polybutyl methacrylate, plasticized polyvinyl chloride, plasticized nylon, plasticized soft nylon, plasticized polyethylene terephthalate, natural rubber, polyisoprene, polyisobutylene, polybutadiene, polyethylene, polytetrafluoroethylene, polyvinylidene chloride, polyacrylonitrile, cross-linked polyvinylpyrrolidone, polytrifluorochloroethylene, chlorinated polyethylene, poly(1,4′-isopropylidene diphenylene carbonate), vinylidene chloride, acrylonitrile copolymer, vinyl chloride-diethyl fumarate copolymer, silicone rubbers, ethylene-propylene rubber, silicone-carbonate copolymers, vinylidene chloride-vinyl chloride copolymer, vinyl chloride-acrylonitrile copolymer, and vinylidene chloride-acrylonitrile copolymer.

8. A sustained release drug delivery system in accordance with any one of claims 1 - 7 , wherein the agent is fluocinolone acetonide.

9. The sustained release drug delivery device in accordance with claim 1 , wherein the device is not a laminate.

10. The sustained release drug delivery device in accordance with claim 1 , wherein the length of the tube is not more than 7 millimeters.

11. A method of manufacturing a sustained drug delivery system suitable for injection into the vitreous of the eye, comprising

positioning a drug core in a permeable tube, said drug core containing a therapeutically effective amount of an active agent, wherein said permeable tube is not more than 3 millimeters in diameter, dimensionally stable, and capable of supporting its own weight;

cross-sectionally cutting the tube into sections; and

forming a permeable outer layer around at least a portion of a section.

12. The method according to claim 11 , wherein the outer layer is formed by dipping, dropping, spraying, or brushing the section with a solution of polymer.
