Abstract

Provided herein, are droplet mixture compositions and systems and methods for forming mixtures of droplets. The system may comprise two or more droplet generation units. Each unit may include at least one first input well, a second input well, and an output well connected to the first and second input wells by channels that form a droplet generator. The combined droplet populations can be mixed, heated, and collected for multiple uses, such as for use as calibration standards for instrument testing and analysis.

Claims (16)

1. A method comprising the steps of:

(a) combining a first droplet population with a second droplet population to form a combined droplet population;

(b) mixing the combined droplet population in a mixing module to produce a mixture in which each of the first droplet population and the second droplet population remains substantially intact;

(c) stabilizing droplets of the first droplet population and droplets of the second droplet population by heating that forms a droplet skin including a skin-forming protein; and

(d) shipping at least a portion of the combined droplet population at least one kilometer after the steps of mixing and stabilizing.

2. The method of claim 1 , wherein the step of mixing the combined droplet population includes a step of flowing the combined droplet population through the mixing module using a pressure source to drive flow of the combined droplet population.

3. The method of claim 2 , wherein the step of flowing the combined droplet population includes a step of flowing the combined droplet population through a mixing channel of the mixing module.

4. The method of claim 3 , wherein the mixing channel has a nonlinear configuration including at least one turn.

5. The method of claim 1 , wherein the step of combining a first droplet population with a second droplet population occurs in the mixing module.

6. The method of claim 1 , further comprising a step of detecting a signal from the combined droplet population.

7. The method of claim 6 , wherein droplets of the combined droplet population have been stored at least 24 hours before the step of detecting a signal.

8. The method of claim 6 , wherein the step of detecting a signal includes a step of detecting a relatively high fluorescence emission from droplets of the first droplet population and a relatively low fluorescence emission from droplets of the second droplet population.

The method of claim 1 , further comprising a step of partitioning a sample with a first droplet generator to produce the first droplet population and a step of partitioning a sample with a second droplet generator to produce the second droplet population, wherein the first droplet generator is different from the second droplet generator.

The method of claim 1 , wherein, during the step of combining a first droplet population with a second droplet population, at least a portion of the first droplet population is present in a first channel and at least a portion of the second droplet population is present in a second channel that is different from the first channel.

11. The method of claim 10 , wherein the first channel and the second channel intersect one another.

12. The method of claim 10 , wherein the first channel and the second channel are in fluid communication with the mixing module during the step of combining a first droplet population with a second droplet population.
