Abstract

The invention features devices and methods for the deterministic separation of particles. Exemplary methods include the enrichment of a sample in a desired particle or the alteration of a desired particle in the device. The devices and methods are advantageously employed to enrich for rare cells, e.g., fetal cells, present in a sample, e.g., maternal blood and rare cell components, e.g., fetal cell nuclei. The invention further provides a method for preferentially lysing cells of interest in a sample, e.g., to extract clinical information from a cellular component, e.g., a nucleus, of the cells of interest. In general, the method employs differential lysis between the cells of interest and other cells (e.g., other nucleated cells) in the sample.

Claims (41)

1. A device comprising:

a channel extending from one or more inlets to one or more outlets, wherein the channel is bounded by a first wall and a second wall located opposite from the first wall;

a first array of obstacles disposed within the channel and bounded by the first wall and a first array boundary;

a second array of obstacles disposed within the channel and bounded by the second wall and a second array boundary; and

a bypass channel defined by a region devoid of obstacles between the first array boundary and the second array boundary,

wherein the first array and the second array are configured to deflect particles in a fluid flowed through the first array and the second array, respectively, toward the bypass channel.

The device of claim 1 , wherein the first array of obstacles comprises a plurality of first obstacles that form a network of gaps and comprises at least first and second rows of the obstacles, wherein the second row is displaced laterally relative to the first row so that a fluid passing through a gap in the first row is divided unequally when flowing around an obstacle and into two gaps on either side of the obstacle in the second row.

3. The device of claim 1 , wherein the one or more inlets further comprise:

a first inlet located closer to the first wall than to the bypass channel to receive the fluid for flowing through the first array of structures; and

a second inlet located closer to the second wall than to the bypass channel to receive the fluid for flowing through the second array of structures.

4. The device of claim 1 , wherein the one or more outlets further comprise:

a first outlet located closer to the first wall than to the bypass channel to receive particles not deflected by the first array;

a second outlet located closer to the second wall than to the bypass channel to receive particles not deflected by the second array; and

a third outlet located at an end of the bypass channel to receive particles deflected by the first array and the second array and collected in the bypass channel.

The device of claim 2 , wherein the second array of structures comprises a plurality of second obstacles that form a network of gaps and comprises at least first and second rows of the obstacles, wherein the second row is displaced laterally relative to the first row so that fluid passing through a gap in the first row is divided unequally when flowing around an obstacle and into two gaps in the second row.

6. The device of claim 1 , wherein the bypass channel is disposed along a longitudinal axis of the channel.

7. The device of claim 1 , further comprising:

a third array of structures disposed within the channel in series with the first array of structures, and bounded by the first wall and a third array boundary; and

a fourth array of structures disposed within the channel in series with the second array of structures, and bounded by the second wall and a fourth array boundary,

wherein the bypass channel extends to a region between the third array boundary and the fourth array boundary.

8. The device of claim 1 wherein the array of obstacles form a network of gaps of a substantially constant width of 10-30 μm.

9. The device of claim 8 wherein the width of said gaps is 18-24 μm.

10. The device of claim 9 , wherein the obstacles are shaped with a maximum dimension in an average flow direction of the array that is less than a maximum dimension orthogonal to the average flow direction of the array.

11. The device of claim 7 , further comprising:

a second bypass channel defined by a region between the third array boundary and the bypass channel; and

a third bypass channel defined by a region between the fourth array boundary and the bypass channel.

12. The device of claim 1 , wherein the first array and the second array are disposed on either side of a longitudinal axis of the channel.

13. The device of claim 11 , wherein the first array is a mirror image of the second array.

14. The device of claim 1 , further comprising fin structures adjacent the bypass channel and the first array to prevent fluid that has flowed into the bypass channel from re-entering the first array.

15. A device comprising:

a channel extending from one or more inlets to one or more outlets, wherein the channel is bounded by a first wall and a second wall located opposite from the first wall;

a first plurality of rows of obstacles disposed within the channel, wherein each row of the first plurality extends from the first wall towards the second wall, wherein ends of the first plurality of rows adjacent the second wall define a first boundary; and

a first bypass channel defined by a first region devoid of obstacles between the first boundary and the second wall;

wherein the first array is configured to deflect particles in a fluid flowed through the first array toward the first bypass channel.

16. The device of claim 15 , further comprising:

a second plurality of rows of obstacles disposed within the channel in series with the first plurality of rows, wherein each row of the second plurality extends from the first wall towards the second wall, wherein ends of the second plurality adjacent the second wall define a second boundary; and

a second bypass channel defined by a second region devoid of obstacles between the second boundary and the second wall, wherein the second bypass channel is wider than the first bypass channel.

The device of claim 16 , wherein each of the first plurality and the second plurality of rows includes respective first and second rows, wherein the second row is displaced laterally relative to the first row so that a fluid passing through a gap in the first row is divided unequally when flowing around an obstacle and into two gaps on either side of an obstacle in the second row, and wherein a gap between first and second rows in the first plurality is larger than a gap between first and second rows in the second plurality.

18. The device of claim 15 , wherein a width of the first bypass channel gradually increases along an axial direction of fluid flow.

19. The device of claim 15 , wherein a width of the first bypass channel gradually decreases along an axial direction of fluid flow.

20. The device of claim 15 , wherein the first bypass channel is designed to remove an amount of flow through the first plurality of rows so that the fluid flow in the first plurality of rows is not perturbed.
