Abstract

System and method for fluorescent light excitation and detection from samples to enhance the numerical aperture and/or reduce the cross-talk of the fluorescent light.

Claims (30)

1 . A method for analyzing samples comprising:

directing excitation light into at least one housing configured to contain samples, wherein the excitation light is propagated within the housing by total internal reflection;

introducing the excitation light into the at least one housing through a wall of the at least one housing; and

collecting sample emissions using at least one NA enhancing optical element which has an index of refraction greater than the housing.

2 . The method of claim 1 , further comprising:

selectively bounding excitation light and sample emissions using a mask having an aperture.

3 . The method of claim 2 , wherein selective bounding of the excitation light and sample emissions by the mask is adapted to reduce cross-talk between two or more housings.

4 . The method of claim 1 , wherein the NA enhancing optical element is formed as a truncated sphere.

5 . The method of claim 1 , wherein excitation light introduced into the housing passes through a coupling optical element associated with the wall of the housing.

6 . The method of claim 5 , wherein the coupling optical element has an index of refraction greater than an index of refraction of the housing.

7 . The method of claim 1 , wherein a second excitation light is directed into the at least one housing passing through a second coupling optical element.

8 . The method of claim 7 , wherein the second coupling optical element has an index of refraction greater than an index of refraction of the housing.

9 . An sample illumination apparatus comprising:

a housing having an associated coupling optical element which receives excitation light and propagates the excitation light through the housing by total internal reflection wherein the coupling optical element has an index of refraction greater than that of the housing and whereby at least one sample contained in the housing is illuminated by the excitation light and emits a signal in response.

10 . The apparatus of claim 9 further comprising:

at least one NA enhancing optical element which has an index of refraction greater than the housing through which sample emissions pass.

11 . The apparatus of claim 9 , wherein the housing comprises a plurality of capillaries.

12 . The apparatus of claim 9 , further comprising:

a mask having an aperture for selectively bounding excitation light and sample emissions.

13 . The apparatus of claim 12 , wherein the mask selective bounds excitation light or sample emissions between two or more housings.

14 . A method for analyzing samples comprising:

directing non-coherent light into a housing configured to transport samples and propagate light wherein the housing is associated with a coupling optical element which introduces non-coherent light into the housing through a wall of the housing and wherein the coupling optical element has an index of refraction greater than an index of refraction of the housing; and

detecting sample emissions resulting from the non-coherent light interacting with the samples transported in the housing.

15 . The method of claim 14 , wherein the housing comprises a plurality of capillaries.

16 . The method of claim 14 further comprising:

focusing the non-coherent light using at least one high NA optical element.

17 . The method of claim 16 , wherein the at least one high NA optical element is formed as a truncated sphere.

18 . The method of claim 16 , wherein the at least one high NA optical element is formed as a meniscus lens.

19 . The method of claim 14 , wherein the housing is formed as a capillary and the sample is fluidically transported within the capillary, wherein the sample fluid has an index of refraction greater than an index outside of the housing.

20 . The method of claim 14 , wherein sample emissions are detected at a detection zone axially illuminated by the non-coherent light introduced into the housing.
