Abstract

Oral compositions and methods of use thereof are provided herein. The oral compositions comprise a first component comprising at least one E h -raising compound and a pharmaceutically acceptable carrier, and a second component comprising at least one zinc compound, an anti-microbial agent and a pharmaceutically acceptable carrier.

Claims (45)

1. An oral composition comprising:

a first component comprising at least one E h -raising compound and a pharmaceutically acceptable carrier, wherein the E h -raising compound is sodium chlorite or sodium bromite and

a second component comprising at least one zinc compound capable of providing freely available zinc ions, cetylpyridinium chloride (CPC) and a pharmaceutically acceptable carrier;

wherein the first component comprises about 0.01% by weight to about 3.0% by weight of the at least one E h -raising compound, and wherein the second component comprises about 0.02% by weight to about 1.0% by weight of zinc compound and about 0.02% by weight to about 0.6% by weight of CPC; and

wherein the oral composition is effective to maintain oral volatile sulfur compound concentrations below 350 ppb for at least six hours after rinse.

2. The oral composition of claim 1 , wherein the first and second components are stored separately.

3. The oral composition of claim 1 , wherein the first component further comprises a chlorine-containing compound.

4. The oral composition of claim 3 , wherein the chlorine-containing compound is selected from the group consisting of alkali metal chloride salts, alkaline earth metal chloride salts, and combinations thereof.

5. The oral composition of claim 1 , wherein the at least one zinc compound is selected from the group consisting of zinc chloride, zinc acetate, zinc salicylate, zinc sulfate, zinc nitrate, and combinations thereof.

6. The oral composition of claim 1 , wherein the at least one zinc compound is selected from the group consisting of zinc chloride and zinc acetate.

The oral composition of claim 4 , wherein the first component comprises about 0.5% by weight to about 2.5% by weight of the chlorine-containing compound, and wherein the second component comprises about 0.02% by weight to about 1.0% by weight of zinc compound and about 0.02% by weight to about 0.6% by weight of CPC.

8. The oral composition of claim 1 , wherein a pH of the first component ranges from about 7.0 to about 8.5, and a pH of the second component ranges from about 3.0 to about 6.0.

9. The oral composition of claim 1 , further comprising essential oils.

10. The oral composition of claim 1 , further comprising xylitol.

11. The oral composition of claim 1 , wherein the oral composition further comprises at least one further component selected from the group consisting of a desensitizing agent, a whitening agent, an anti-microbial agent, an antibiotic, an anti-cavity agent, an anti-plaque agent, an anti-tartar agent, an agent to alleviate or reduce dry mouth, a deodorizer, a polishing agent, a detergent and a sweetener.

The oral care composition of claim 11 , wherein the desensitizing agent comprises at least one of benzocaine, potassium nitrate, a fluoride salt, strontium chloride, potassium chloride, dibasic sodium citrate, iron oxalate, aluminum lactate, sodium nitrate, lithium nitrate, magnesium nitrate, calcium nitrate, sodium citrate, dibasic sodium citrate in a poloxamer gel, potassium tartrate, calcium hydroxide, dibasic calcium phosphate, strontium acetate, sodium monofluorophosphate, formaldehyde, bisabolol, aspirin, ibuprofen, codeine, acetaminophen, sodium salicylate, and triethanolamine salicylate and combinations thereof.

13. The oral composition of claim 11 , wherein the whitening agent comprises at least one of carbamide peroxide, hydrogen peroxide, sodium bicarbonate, and combinations thereof.

14. The oral composition of claim 11 , wherein the anti-cavity agent comprises at least one of fluoride, sodium fluoride, sodium monofluorophosphate, nicometanol fluorhydrate, ammonium fluoride, and potassium fluoride.

The oral composition of claim 11 , wherein the anti-plaque agent comprises at least one of an alcohol, triclosan, sanguinarine, hexetidyne, zinc citrate, a fluoride, lauryl sodium sulfate, zinc phosphate, zinc acetate, zinc aspartate, zinc acetylmethionate, zinc citrate trihydrate, zinc tannate, zinc gluconate, zinc lactobionate, zinc maltobionate, zinc hydrolyzed collagen, zinc pyrrolidone carboxylic acid (zinc PCA), zinc tribromosalicylanfiide, zinc caprylate, zinc octoate, zinc laurate, zinc myristate, zinc stearate, zinc oleate, zinc carbonate, zinc borate, zinc silicate, zinc sulfide, zinc sulfate, zinc oxide, zinc phenol sulfonate, zinc stannate, zinc dl-lactate, trihydrate, zinc cocoate, tannic acid, citric acid, acetic acid, lactic acid, sodium trihydrogen pyrophosphate, disodium dihydrogen pyrophosphate, trisodium hydrogen pyrophosphate, trisodium hydrogen pyrophosphate monohydrate, trisodium hydrogen pyrophosphate nonahydrate, tetrasodium pyrophosphate, tetrasodium pyrophosphate decahydrate, potassium trihydrogen pyrophosphate, dipotassium dihydrogen pyrophosphate, tripotassium hydrogen pyrophosphate, tetrapotassium pyrophosphate, diammonium dihydrogen pyrophosphate, triammonium hydrogen pyrophosphate, triammonium hydrogen pyrophosphate monohydrate, calcium dihydrogen pyrophosphate, calcium pyrophosphate, tetraaluminium pyrophosphate, dextranase, mutanase, cellulase, papain, bromelin and combinations thereof.

16. The oral composition of claim 11 , wherein the anti-tartar agent comprises at least one of pyrophosphate, a zinc salt and combinations thereof.

17. The oral composition of claim 11 , wherein the agent to alleviate or reduce dry mouth comprises at least one of lactoferrin, lysozyme, lactoperoxidase, immunoglobulins, colustrum extract, glucose oxidase, amylase, amyloglucosidase, glucoxidase, papain, peptizyme and aloe vera.

18. The oral composition of claim 11 , wherein the deodorizer comprises at least one of chlorhexidine, hydrogen peroxide, vitamin B, vitamin C, sodium bicarbonate, an herb, rue and combinations thereof.

The oral composition of claim 11 , wherein the polishing agent comprises at least one of calcium carbonate, dicalcium phosphate dihydrate, aluminum hydroxide, dental grade silicas, precipitated calcium carbonate, light calcium carbonate, medium calcium carbonate, dense calcium carbonate, extra dense calcium carbonate, sodium carbonate, sodium bicarbonate, potassium carbonate, potassium bicarbonate, anhydrous calcium phosphate, dicalcium phosphate, tribasic calcium phosphate, sodium phosphate, potassium phosphate, calcium pyrophosphate, insoluble sodium metaphosphate, disodium orthophosphate, dibasic sodium phosphate, magnesium hydroxide, magnesium carbonate, magnesium silicate, magnesium trisilicate, trimagnesium phosphate, monomagnesium phosphate, magnesium oxide, stannic oxide, zinc oxide, chalk, extra light chalk, light chalk, medium chalk, course chalk, bentonite, pumice flour, α-alumina trihydrate, levigated alumina, hydrated alumina, alumina aerogel, gelatinous alumina, aluminum silicate, calcined aluminum silicate, zirconium silicate, hydroxyapatite, crosslinked urea-formaldehyde resin, crosslinked melamine-formaldehyde resin, polymethacrylate, polymethylmethacrylate, polystyrene, powdered polyethylene, silica gel, dehydrated silica gel, silica precipitates, silica xerogel, silica hydrogels, silica pyrogel, silica aerogel, sodium glycerophosphate, sodium trimetaphosphate, organic phosphates and combinations thereof.

20. The oral composition of claim 11 , wherein the detergent comprises at least one of sodium lauryl sulphate, sodium lauryl sarcosinate, sodium methyl cocoyl taurate, polysorbates, sorbitan derivatives and combinations thereof.

The oral composition of claim 1 , wherein the first component comprises about 0.04% by weight to about 1.2% by weight of sodium chlorite and a pH of the first component ranges from about 7.0 to about 8.5, and wherein the second component comprises about 0.04% by weight to about 0.7% by weight of zinc compound and about 0.09% by weight to about 0.2% by weight of CPC and a pH of the second component ranges from about 3.0 to about 6.0.

22. The oral composition of claim 2 , wherein the first and the second components are stored separately in one or more containers, a syringe, a two-phase tablet or chew.

23. The oral composition of claim 1 , wherein the first component is a solution and the second component is a solution.

24. The oral composition of claim 1 , wherein one of the components is a solution and the other component is present in an encapsulated form within the solution.

25. The oral composition of claim 1 , wherein the composition is a solid, liquid, spray, gel, foam, syrup, or powder.

26. The oral composition of claim 1 , wherein the oral composition is in a form selected from the group consisting of a dental care product, a food product, a tablet, a flash-melt formulation, a candy, a lozenge, a chewing gum, a confection, a toothpaste, a mouthrinse, a breath spray and a mint.

27. The oral composition of claim 1 comprising:

about 0.005% by weight to about 1.5% by weight of E h -raising compound; about 0.01% by weight to about 0.5% by weight of zinc compound; about 0.01% by weight to about 0.3% by weight of CPC; and at least one pharmaceutically acceptable carrier, wherein the oral composition is prepared by mixing a first component comprising the E h -raising compound and a pharmaceutically acceptable carrier and a second component comprising a zinc compound, CPC and a pharmaceutically acceptable carrier.

28. The oral composition of claim 1 , wherein the oral composition is effective to maintain oral volatile sulfur compound concentrations below 350 ppb for at least eight hours after rinse.

29. A method for reducing oral malodor comprising delivering the oral composition of claim 1 to an oral cavity in a subject.

30. The method of claim 29 , further comprising, prior to delivering the oral composition to an oral cavity in a subject, mixing the first component and the second component.

31. The method of claim 29 , wherein the method is effective to reduce oral malodor from about 2 hours to about 12 hours.

32. A method for treating or alleviating a dental disease comprising delivering the oral composition of claim 1 to an oral cavity in a subject.

33. The method of claim 32 , further comprising, prior to delivering the oral composition to an oral cavity in a subject, mixing the first component and the second component.

34. The method of claim 32 , wherein the dental disease is selected from the group consisting of gingivitis, periodontitis and tooth decay.

35. A method for reducing plaque comprising delivering the oral composition of claim 1 to an oral cavity in a subject.

36. The method of claim 35 , further comprising, prior to delivering the oral composition to an oral cavity in a subject, mixing the first component and the second component.

37. A method to reduce canker sores comprising delivering the oral composition of claim 1 to an oral cavity in a subject.

38. The method of claim 37 , further comprising, prior to delivering the oral composition to an oral cavity in a subject, mixing the first component and the second component.

39. The method of claim 29 , wherein the subject is human.

40. The method of claim 29 , wherein the subject is a non-human animal.
