Abstract

Proteases such as memapsin-1 are import enzymes, playing roles in a variety of diseases including diabetes. The inventors have developed inhibitors of memapsin 1 and methods of use therefore in the treatment of disease.

Claims (56)

1. A method of inhibiting memapsin 1 activity comprising contacting a memapsin 1 enzyme with a compound having a formula:

wherein R 5 is:

wherein:

R 9 is hydrogen or halo;

R 10 is alkyl (C≦4) ;

R 6 is hydrogen, alkyl (C≦8) , aralkyl (C≦8) , or a substituted version of the alkyl (c≦8) or aralkyl (C≦8) groups; or a halogen; and

R 7 is:

R 8 is hydrogen, alkyl (C≦8) , or

wherein: R 12 and R 13 are each independently hydrogen, alkyl (C≦8) , or alkylsulfonyl (C≦8) ,

wherein (i) if R 9 is hydrogen, then R 8 must be hydrogen or alkyl (C≦8) , and (ii) if R 8 is

 then R 9 must be halo, and one or both of R 12 and R 13 must be hydrogen,

or a pharmaceutically acceptable salt or tautomer.

2. The method of claim 1 , wherein the compound has formula III, wherein the formula is:

wherein:

R 9 is hydrogen or halo;

R 6 is hydrogen, alkyl (C≦8) , aralkyl (C≦8) , or a substituted version of the alkyl (C≦8) or aralkyl (C≦8) groups; or a halogen; and

R 7 is:

or a pharmaceutically acceptable salt or tautomer.

3. The method of claim 1 , wherein the compound has formula III, wherein R 5 is

4. The method of claim 3 , wherein R 9 is hydrogen.

5. The method of claim 3 , wherein R 9 is halo.

6. The method of claim 3 , wherein R 10 is methyl.

7. The method of claim 1 , wherein the compound has formula III, wherein R 6 is benzyl or isobutyl.

8. The method of claim 1 , wherein the compound has formula III, wherein R 7 is:

9. The method of claim 1 , wherein the compound has formula III, wherein R 8 is hydrogen.

10. The method of claim 1 , wherein the compound has formula III, wherein R 8 is alkyl (C≦8) .

11. The method of claim 1 , wherein the compound has formula III, wherein R 8 is

12. The method of claim 11 , wherein one of R 12 and R 13 is alkyl (C≦8) .

13. The method of claim 11 , wherein one of R 12 and R 13 is methyl.

14. The method of claim 11 , wherein one of R 12 and R 13 is alkylsulfonyl (C≦8) .

15. The method of claim 14 , wherein one of R 12 and R 13 is methylsulfonyl.

16. The method of claim 1 , wherein the compound has formula III, wherein R 9 is iodo.

17. A method of treating a mammalian subject with type 2 diabetes comprising administering to said subject a compound having a formula:

wherein R 5 is:

wherein:

R 9 is hydrogen or halo;

R 10 is alkyl (C≦4) ;

R 6 is hydrogen, alkyl (C≦8) , aralkyl (C≦8) , or a substituted version of the alkyl (C≦8) or aralkyl (C≦8) groups; or a halogen; and

R 7 is:

R 8 is hydrogen, alkyl (C≦8) , or

wherein: R 12 and R 13 are each independently hydrogen, alkyl (C≦8) , or alkylsulfonyl (C≦8) ,

wherein (i) if R 9 is hydrogen, then R 8 must be hydrogen or alkyl (C≦8) , and (ii) if R 8 is

 then R 9 must be halo, and one or both of R 12 and R 13 must be hydrogen,

or a pharmaceutically acceptable salt or tautomer.

18. A method of increasing pancreatic beta cell mass in a mammalian subject comprising administering to said subject a compound having a formula:

wherein R 5 :

wherein:

R 9 is hydrogen or halo;

R 10 is alkyl (C≦4) ;

R 6 is hydrogen, alkyl (C≦8) , aralkyl (C≦8) , or a substituted version of the alkyl (C≦8) or aralkyl (C≦8) groups; or a halogen; and

R 7 is:

R 8 is hydrogen, alkyl (C≦8) , or

wherein: R 12 and R 13 are each independently hydrogen, alkyl (C≦8) , or alkylsulfonyl (C≦8) ,

wherein (i) if R 9 is hydrogen, then R 8 must be hydrogen or alkyl (C≦8) , and (ii) if R 8 is

 then R 9 must be halo, and one or both of R 12 and R 13 must be hydrogen,

or a pharmaceutically acceptable salt or tautomer.
