Abstract

An altered antibody chain is produced in which the CDR's of the variable domain of the chain are derived from a first mammalian species. The framework-encoding regions of DNA encoding the variable domain of the first species are mutated so that the mutated framework-encoding regions encode a framework derived from a second different mammalian species. The or each constant domain of the antibody chain, if present, are also derived from the second mammalian species. An antibody which is capable of binding to human CD4 antigen is also provided together with a pharmaceutical composition comprising the antibody.

Claims (24)

1. A process for the preparation of an antibody chain having a variable domain comprising three complementarity determining region(s) (CDRs), designated CDR1, CDR2 and CDR3, and a framework, wherein the CDRs are those of a first mammalian species and the framework and any constant domain present are those of a second different mammalian species, which process comprises:

(i) mutating one or more individual bases within the framework-encoding regions of DNA encoding a variable domain of an antibody chain of the said first mammalian species such that the resulting framework-encoding regions encode the said framework of the said second mammalian species; and

(ii) expressing the said antibody chain utilising the DNA resulting from step (i);

step (i) being such that an antibody incorporating the antibody chain expressed in step (ii) retains the antigen binding capability of the antibody of the first mammalian species,

wherein the variable domain framework of said second mammalian species is selected from variable domain sequences of the second mammalian species that have the most overall homology to that of said variable domain sequence of the first mammalian species, homology being determined on the basis of the respective variable domains including at least CDR1 and CDR2.

2. A process according to claim 1 wherein the DNA of step (i) is DNA encoding the variable domain of an antibody heavy chain.

3. A process according to claim 1 , wherein the DNA of step (i) is DNA encoding the variable domain of an antibody light chain.

4. A process according to claim 1 wherein the said first mammalian species is rat or mouse.

5. A process according to claim 1 wherein the said second mammalian species is human.

6. A process according to claim 5 wherein the said first mammalian species is rat or mouse.

7. A process according to claim 1 , wherein the said antibody chain is co-expressed with a complementary antibody chain and antibody comprising the two chains is recovered.

8. A process as claimed in claim 1 , wherein the variable domain framework of the second mammalian species is that selected from variable domains having CDR1 and CDR2 of the same length as that of the CDR 1 and CDR 2 of the variable domain framework of the first mammalian species.

9. A process as claimed in claim 1 wherein the variable domain framework of the second mammalian species is selected by determining homology between the entirety of the two variable domain sequences including CDR1, CDR2 and CDR3.

A process for the preparation of an antibody chain having a variable domain comprising three complementarity determining region(s) (CDRs), designated CDR1, CDR2 and CDR3, and a framework, wherein the CDRs are those of a first mammalian species and the framework and any constant domain present are those of a second different mammalian species, which process comprises:

(i) mutating one or more individual bases within the framework-encoding regions of DNA encoding a variable domain of an antibody chain of said first mammalian species such that the resulting framework-encoding regions encode said framework of the second mammalian species; and

(ii) expressing said antibody chain utilizing the DNA resulting from step (i), step (i) being such that an antibody incorporating the antibody chain expressed in step (ii) retains the antigen binding capability of the antibody of the first mammalian species, said process comprising:

(a) determining the nucleotide and predicted amino acid sequence of a variable domain of a selected antibody chain of the first mammalian species;

(b) determining the antibody framework to which the framework of said domain is to be altered by selecting an antibody chain of said second different mammalian species on the basis of homology between the variable domains of the two antibody chains;

(c) mutating one or more individual bases within the framework-encoding regions of DNA encoding said variable domain such that the resulting framework-encoding regions encode the framework determined upon in step (b);

(d) linking the DNA resulting from step (c) to DNA encoding a constant domain of said second mammalian species and cloning the DNA into an expression vector; and

(e) introducing the expression vector into a compatible host and culturing the host cell under such conditions that the antibody chain is expressed,

wherein the antibody framework region to which the framework is to be altered is selected as that of a variable domain sequence of the second mammalian species which has highest homology to that of the first species, homology being determined on the basis of respective variable domains including at least CDR1 and CDR2.

11. A process as claimed in claim 10 wherein the selected framework region is that selected as having highest homology to the variable domain of the first mammalian species from sequences where CDR1 and CDR2 are of the same length.

A process as claimed in claim 10 wherein the variable domain framework of the second mammalian species is selected as that of the variable domain having the highest homology to the variable domain of the first mammalian species wherein the entirety of the two variable domain sequences is compared including CDR1, CDR2 and CDR3.
