Abstract

Amino acid sequences and corresponding nucleic acid sequence of retinoid metabolizing protein found in human, mouse and zebrafish are described, as well as methods of using same.

Claims (11)

1. A microsomal preparation of a cell that has been transfected with a nucleic acid molecule encoding a protein, or of a desendent cell thereof, wherein said protein oxidizes all-trans retinoic acid at the C4-position of the β-ionone ring, said nucleic acid molecule comprising a nucleotide sequence that hybridizes under high strigency conditions, wherein high stringency conditions include a wash step of about 0.2×SCC at 65° C., to a polynucleotide having a nucleotide sequence shown as SEQ ID NO:3, said microsomal preparation comprising said protein.

A microsomal preparation of a cell that has been transfected with a nucleic acid molecule encoding a protein, or of a desendent cell thereof, wherein said protein hydroxylates all-trans retinoic acid at the C4-position of the β-ionone ring, said nucleic acid molecule comprising a nucleotide sequence that hybridizes under high strigency conditions, wherein high stringency conditions include a wash step of about 0.2×SCC at 65° C., to a nucleic acid molecule having a nucleotide sequence shown as SEQ ID NO:3, said microsomal preparation comprising said protein.

3. The preparation of claim 1 , wherein the protein comprises the amino acid sequence identified as SEQ ID NO:2.

4. The preparation of claim 1 , wherein the nucleic acid molecule encodes an amino acid sequence that is at least 95 percent conserved with respect to SEQ ID NO:2.

5. The preparation of claim 1 , wherein the protein additionally hydroxylates the C18-position of all-trans retinoic acid.

6. The preparation of claim 2 , wherein the nucleic acid molecule encodes an amino acid sequence that is at least 95 percent conserved with respect to SEQ ID NO:2.

7. The preparation of claim 2 , wherein the protein additionally hydroxylates the C18-position of all-trans retinoic acid.

8. A microsomal preparation of a cell that has been transfected with a nucleic acid molecule encoding a protein, or of a desendent cell thereof, wherein said protein oxidizes all-trans retinoic acid at the C4-position of the β-ionone ring, said nucleic acid molecule encoding an amino acid sequence that is at least 95 percent conserved with respect to SEQ ID NO:2, said microsomal preparation comprising said protein.

9. The preparation of claim 8 , wherein the protein additionally hydroxylates the C18-position of all-trans retinoic acid.

10. A microsomal preparation of a cell that has been transfected with a nucleic acid molecule encoding a protein, or of a desendent cell thereof, wherein said protein hydroxylates all-trans retinoic acid at the C4-position of the β-ionone ring, said nucleic acid molecule encoding an amino acid sequence that is at least 95 percent conserved with respect to SEQ ID NO:2, said microsomal preparation comprising said protein.

11. The preparation of claim 10 , wherein the protein additionally hydroxylates the C18-position of all-trans retinoic acid.
