Abstract

Engineered protein kinases which can utilize modified nucleotide triphosphate substrates that are not as readily utilized by the wild-type forms of those enzymes, and methods of making and using them. Modified nucleotide triphosphate substrates and methods of making and using them. Methods for using such engineered kinases and such modified substrates to identify which protein substrates the kinases act upon, to measure the extent of such action, and to determine if test compounds can modulate such action. Also Engineered forms of multi-substrate enzymes which covalently attach part or all of at least one (donor) substrate to at least one other (recipient) substrate, which engineered forms will accept modified substrates that are not as readily utilized by the wild-type forms of those enzymes. Methods for making and using such engineered enzymes. Modified substrates and methods of making and using them. Methods for using such engineered enzymes and such modified substrates to identify the recipient substrates the enzymes act upon, to measure the extent of such action, and to measure whether test compounds modulate such action.

Claims (26)

1. A radiolabeled compound of the following structural formula that serves as a substrate for a mutant form of a wild-type kinase,

wherein the compound does not serve as a substrate for the wild-type kinase,

wherein X is selected from the group consisting of —NH—O-alkyl, —NH—O-cycloalkyl, —NH—O-cycloalkyl, —NH—C(O)-alkyl, —NH-alkylaryl, —NH—O-alkylaryl, and —N-heterocycloalkyl where the nitrogen atom is part of the heterocycloalkyl ring and

wherein at least one of the phosphorus atoms is radioactive.

2. The compound of claim 1 , wherein the —NH—O-alkyl is —NH—OCH 3 , —NH—OCH 2 CH 3 or —NH—O—CH(CH 3 ) 2 .

3. The compound of claim 1 , wherein the —NH—O-cycloalkyl

4. The compound of claim 1 , wherein the —NH-cycloalkyl

5. The compound of claim 1 , wherein the —N-heterocycloalkyl

or

6. The compound of claim 1 , wherein the —NH—C(O)-alkyl is —NH—C(O)—CH 3 .

7. The compound of claim 1 , wherein the —NH-alkylaryl is —NH—CH 2 —Ph.

8. The compound of claim 1 , wherein the —NH—O-alkylaryl is —NH—O—CH 2 —Ph.

9. A kit comprising at least one compound of claim 1 and a container for using it.

10. A compound of the following structural formula

wherein X is selected from the group consisting of —NH—O-alkyl, —NH—O-cycloalkyl, —NH-cycloalkyl, —NH—C(O)-alkyl, —NH—O-alkylaryl and —NH-heterocycloalkyl where the nitrogen atom is part of the heterocycloalkyl ring.

11. The compound of claim 10 , wherein the —NH—O-alkyl is —NH—OCH 3 , —NH—OCH 2 CH 3 or —NH—O—CH(CH 3 ) 2 .

12. The compound of claim 10 , wherein the —NH—O-cycloalkyl

or

13. The compound of claim 10 , wherein the —NH-cycloalkyl

or

14. The compound of claim 10 , wherein the —N-heterocycloalkyl

or

15. The compound of claim 10 , wherein the —NH—C(O)-alkyl is —NH—C(O)-CH 3 .

16. The compound of claim 10 , wherein the —NH—O-alkylaryl is —NH—O—CH 2 —Ph.

17. The compound of claim 10 , wherein at least one of the phosphorus atoms is radioactive.

18. A kit comprising at least compound of claim 10 and a container for using it.
