Abstract

Stable, viscous, mucoadhesive aqueous compositions which are useful for the prevention and treatment of ulcerative, inflammatory, and/or erosive disorders of mucous membranes and/or the delivery of pharmaceutically active compounds to mucosal surfaces for topical treatment or transfer to the systemic circulation.

Claims (26)

1. A pseudoplastic liquid composition for treating or inhibiting an oral mucocutaneous disorder, said liquid composition having a zero shear viscosity at 37±1 degrees Celsius of 100-20,000 centipoise, comprising 0.1 to 3.0 wt/wt % of one or more mucoadhesives in an aqueous solution, wherein the mucoadhesive(s) is/are one or more linear or cross-linked polymer(s) selected from the group consisting of polyacrylic acid, carbopol, carboxymethylcellulose, hydroxyalkylcellulose, dextran sulfate, chitosan, and a water-soluble vinyl polymer; wherein said liquid composition, when administered to an oral mucosa:

1) remains liquid when shear is applied due to swishing the liquid in the mouth; and

2) attaches to and coats the oral mucosa upon discontinuance of said shear.

2. The pseudoplastic liquid composition of claim 1 , further comprising a drug that is therapeutically active against the oral mucocutaneous disorder.

3. The pseudoplastic liquid composition of claim 1 , wherein the mucoadhesive is carbopol.

4. The pseudoplastic liquid composition of either of claim 1 or claim 2 , further comprising one or more viscosity-inducing agent(s).

5. The pseudoplastic liquid composition of either of claim 1 or claim 2 , wherein the mucocutaneous disorder is mucositis, Bechet's disease, apthous ulcer, bullous pemphigoid, chemical systitis, radiation cystitis, erythema multiforme, esophagitis, interstitial cystitis, oral Lichen planus, pemphigus, radiation proctitis, or ulcerative colitis.

6. A pseudoplastic liquid composition for treating or inhibiting an oral mucocutaneous disorder, said liquid composition having a zero shear viscosity at 37±1 degrees Celsius of 100-20,000 centipoise, comprising 0.35 w/w % of one or more mucoadhesives in an aqueous solution, wherein the mucoadhesive(s) is/are one or more linear or cross-linked polymer(s) selected from the group consisting of polyacrylic acid, carbopol, carboxymethylcellulose, hydroxyalkylcellulose, dextran sulfate, chitosan, and a water-soluble vinyl polymer; wherein said liquid composition, when administered to an oral mucosa:

1) remains liquid when shear is applied due to swishing the liquid in the mouth; and

2) attaches to and coats the oral mucosa upon discontinuance of said shear.

7. A pseudoplastic liquid composition for treating or inhibiting an oral mucocutaneous disorder, said liquid composition having a zero shear viscosity at 37±1 degrees Celsius of 100-20,000 centipoise, comprising 0.35 w/w % of one or more mucoadhesives and 5 w/w % of a viscosity inducing agent in an aqueous solution; wherein said liquid composition, when administered to an oral mucosa:

1) remains liquid when shear is applied due to swishing the liquid in the mouth; and

2) attaches to and coats the oral mucosa upon discontinuance of said shear.

8. The pseudoplastic liquid composition of either of claim 6 or claim 7 , further comprising a drug that is therapeutically active against the oral mucocutaneous disorder.

9. The pseudoplastic liquid composition of either claim 6 or 7 , wherein the one or more mucoadhesive is carbopol.

10. The pseudoplastic liquid composition of claim 7 , wherein the viscosity inducing agent is glycerin.

11. A pseudoplastic liquid composition for treating or inhibiting an oral mucocutaneous disorder, said liquid composition having a zero shear viscosity at 37±1 degrees Celsius of 100-20,000 centipoise, comprising 0.35 w/w % of cabopol, 1.5 w/w % of benzyl alcohol, 0.4 w/w % of sodium saccharin, and 0.05% w/w % of polysorbate 60 in an aqueous solution; wherein said liquid composition, when administered to an oral mucosa:

1) remains liquid when shear is applied due to swishing the liquid in the mouth; and

2) attaches to and coats the oral mucosa upon discontinuance of said shear.

12. The pseudoplastic liquid composition of claim 11 , further comprising a drug that is therapeutically active against the mucocutaneous disorder.

13. The pseudoplastic liquid composition of claim 11 , wherein the composition further comprises one or more viscosity-inducing agent (s).

14. The pseudoplastic liquid composition of any of claims 1 , 2 , 3 , 6 , 7 , and 10 - 13 , wherein said pseudoplastic liquid composition has a zero shear viscosity at 37±1 degree Celsius of 5,000-20,000 centipoise.

15. The pseudoplastic liquid composition of claim 4 , wherein said pseudoplastic liquid composition has a zero shear viscosity at 37±1 degree Celsius of 5,000-20,0000 centipoise.

16. The pseudoplastic liquid composition of claim 5 , wherein said pseudoplastic liquid composition has a zero shear viscosity at 37±1 degree Celsius of 5,000-20,0000 centipoise.

17. The pseudoplastic liquid composition of claim 8 , wherein said pseudoplastic liquid composition has a zero shear viscosity at 37±1 degree Celsius of 5,000-20,0000 centipoise.

18. The pseudoplastic liquid composition of claim 9 , wherein said pseudoplastic liquid composition has a zero shear viscosity at 37±1 degree Celsius of 5,000-20,0000 centipoise.
