Abstract

The invention provides isolated N-methyl-D-aspartate type 3B (NR3B) polypeptides, functional fragments and peptides, encoding nucleic acid molecules and polynucleotides, and specific antibodies. Also provided are excitatory glycine receptors, containing either NR3B or NR3A polypeptides. Further provided are methods for detecting excitatory glycine receptor ligands, agonists and antagonists. The invention also provides related diagnostic and therapeutic methods.

Claims (23)

1. A method of detecting an excitatory glycine receptor agonist or antagonist, comprising:

(a) contacting an excitatory glycine receptor comprising an NR3B polypeptide, wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:2, 60, 4, 58, 6, 62, or 8, with one or more candidate compounds under conditions suitable for detecting receptor activation; and

(b) detecting a candidate compound that alters said receptor activation,

wherein said compound altering receptor activation is characterized as an excitatory glycine receptor agonist or antagonist.

2. The method of claim 1 , wherein said excitatory glycine receptor comprises said NR3B polypeptide and an NR1 polypeptide.

3. The method of claim 2 , wherein said excitatory glycine receptor further comprises an NR2 polypeptide.

4. The method of claim 2 , wherein said NR1 polypeptide comprises a naturally-occurring human, rat or mouse NR1 amino acid sequence.

5. The method of claim 1 , wherein said excitatory glycine receptor comprises an NR3A polypeptide and an NR1 polypeptide.

6. The method of claim 5 , wherein said excitatory glycine receptor further comprises an NR2 polypeptide.

7. The method of claim 5 , wherein said NR3A polypeptide comprises a naturally-occurring human or rat NR3A amino acid sequence.

8. The method of claim 5 , wherein said NR1 polypeptide comprises a naturally-occurring human, rat or mouse NR1 amino acid sequence.

9. The method of claim 1 , wherein said receptor activation is detected by assaying whole-cell currents.

10. The method of claim 1 , wherein said receptor activation is detected by assaying single-channel currents.

11. The method of claim 1 , wherein said receptor activation is detected by assaying ion fluxes using ion indicators.

12. The method of claim 9 , wherein said cell is a Xenopus oocyte or mammalian cell that expresses said excitatory glycine receptor.

13. The method of claim 1 , wherein said contacting occurs in the presence of glycine.

14. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:2.

15. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:60.

16. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:4.

17. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:58.

18. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:6.

19. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:62.

20. The method of claim 1 , wherein said NR3B polypeptide comprises the amino acid sequence designated SEQ ID NO:8.
