Protease regulator screening assay
A polypeptide containing an anchor region, a protease recognition site, and a detectable signal region can be produced recombinantly and directly attached to a solid support. The polypeptide is useful for screening protease regulators, especially protease inhibitors.
1. A method for screening a compound for its ability to regulate protease activity comprising the steps of:
incubating, in the presence and absence of a test compound:
a protease; and
a polypeptide, wherein the polypeptide comprises (a) an anchor region, (b) a protease recognition site which comprises a protease cleavable peptide bond and at least 3 naturally adjacent amino acids on each side of the cleavable peptide bond, and (c) a detectable signal region, wherein the anchor region is bound to a solid support; and
detecting the signal region bound to the solid support, wherein detecting an amount of the signal region bound to the solid support in the presence of the test compound which is greater or smaller than an amount detected in the absence of the test compound indicates that the test compound can regulate protease activity.
2. The method of claim 1 wherein the anchor region specifically binds to an antibody.
3. The method of claim 2 wherein the anchor region comprises protein A.
4. The method of claim 1 wherein the protease recognition site is a hepatitis C virus protease recognition site.
5. The method of claim 4 wherein the hepatitis C virus protease recognition site is selected from the group consisting of 4A/4B, 4B/5A, 5A/5B, and serial combinations thereof.
6. The method of claim 1 wherein the detectable signal region comprises an epitope.
7. The method of claim 6 wherein the epitope comprises the amino acid sequence FLAG (SEQ ID NO:1).
8. The method of claim 1 wherein the polypeptide further comprises a linker region between the anchor region and the protease recognition site.
9. The method of claim 1 wherein the anchor region specifically binds to an antibody, the protease recognition site is a hepatitis C virus protease recognition site which is selected from the group consisting of 4A/4B, 4B5A, 5A/5B, and serial combinations thereof, and the signal region comprises an epitope.
10. The method of claim 9 wherein the anchor region comprises protein A and the epitope comprises the amino acid sequence FLAG (SEQ ID NO:1).
11. The method of claim 10 further comprising a linker region between the anchor region and the protease recognition site.
