Method of screening for inhibitors of osteopontin
Amino acids (136 to 142) and amino acids (162 to 168) of human osteopontin constitute two sites at which osteopontin interacts with α4 integrins. Products capable of disrupting those interactions are useful in therapy, particularly in the treatment of inflammatory diseases.
1. A method for identifying a product which is capable of disrupting an interaction between amino acids 162 to 168 of human osteopontin (OPN) and an α4 integrin, which method comprises:
(i) providing, as a first component, a polypeptide which comprises the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4) and RADSVVYGLR (SEQ ID NO: 19);
(ii) providing, as a second component, an α4 integrin;
(iii) contacting the two components under conditions that would permit the two components to interact and determining the degree of interaction between the two components
(iv) contacting the two components with a test product under conditions that would permit the two components to interact in the absence of the test product and determining the degree of interaction between the two components; and
(v) determining whether the test product is capable of disrupting the interaction between the first and second components by comparing the degrees of interaction found in (iii) and (iv).
2. A method according to claim 1 , wherein the α4 integrin used as a second component is α4β1.
3. A method for identifying a product which is capable of binding to amino acids 162 to 168 of human OPN, which method comprises:
(i) providing a first component that is a polypeptide which comprises of the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4) and RADSVVYGLR (SEQ ID NO: 19);
(ii) contacting the first component with an α4 integrin under conditions that, in the absence of a test product, would lead to an interaction between the first component and the α4 integrin;
(iii) contacting the first component with an α4 integrin and a test product under the same conditions that lead to an interaction between the first component and the α4 integrin in (ii); and
(iv) determining whether the test product is capable of binding to the first component.
4. A test kit suitable for use in identifying a product which is capable of disrupting an interaction between amino acids 162 to 168 of human OPN and an α4integrin, which kit comprises:
(i) a first component that is a polypeptide which comprises the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4) and RADSVVYGLR (SEQ ID NO: 19); and
(ii) an α4 integrin.
5. A test kit according to claim 4 , wherein the α4 integrin is α4β1.
6. A polypeptide comprising the amino acid sequence RADSVVYGLR (SEQ ID NO: 19).
7. A pharmaceutical composition comprising a pharmaceutically acceptable carrier or diluent and, as active ingredient, a polypeptide comprising the amino acid sequence RADSVVYGLR (SEQ ID NO: 19).
8. A method for identifying a product which is capable of disrupting an interaction between amino acids 162 to 168 of human osteopontin (OPN) and an α4integrin, which method comprises:
(i) providing, as a first component, a polypeptide which consists of the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4), RGDSVVYGLR (SEQ ID NO: 41) and RADSVVYGLR (SEO ID NO: 19);
(ii) providing, as a second component, an α4 integrin;
(iii) contacting the two components under conditions that would permit the two components to interact and determining the degree of interaction between the two components
(iv) contacting the two components with a test product under conditions that would permit the two components to interact in the absence of the test product and determining the degree of interaction between the two components; and
(v) determining whether the test product is capable of disrupting the interaction between the first and second components by comparing the degrees of interaction found in (iii) and (iv).
9. A method according to claim 8 , wherein the α4 integrin used as a second component is α4β1.
10. A method for identifying a product which is capable of binding to amino acids 162 to 168 of human OPN, which method comprises:
(i) providing a first component that is a polypeptide which consists of the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4), RGDSVVYGLR (SEQ ID NO: 41) and RADSVVYGLR (SEQ ID NO: 19);
(ii) contacting the first component with an α4 integrin under conditions that, in the absence of a test product, would lead to an interaction between the first component and the α4 integrin;
(iii) contacting the first component with an α4 integrin and a test product under the same conditions that lead to an interaction between the first component and the α4 integrin in (ii); and
(iv) determining whether the test product is capable of binding to the first component.
11. A test kit suitable for use in identifying a product which is capable of disrupting an interaction between amino acids 162 to 168 of human OPN and an α4integrin, which kit comprises:
(i) a first component that is a polypeptide which consists of the amino acid sequence selected from the group consisting of: SVVYGLR (SEQ ID NO: 4), RGDSVVYGLR (SEQ ID NO: 41) and RADSVVYGLR (SEQ ID NO: 19); and
(ii) an α4 integrin.
12. A test kit according to claim 11 , wherein the α4 integrin is α4β1.
13. A polypeptide consisting of the amino acid sequence selected from the group consisting of: RGDSVVYGLR (SEQ ID NO: 41) and RADSVVYGLR (SEQ ID NO: 19).
14. A pharmaceutical composition comprising a pharmaceutically acceptable carrier or diluent and, as active ingredient, a polypeptide consisting of the amino acid sequence selected from the group consisting of: RGDSVVYGLR (SEQ ID NO: 41) and RADSVVYGLR (SEO ID NO: 19).
