Abstract

The present invention is directed to the novel use of a CSBP/p38 inhibitor for the treatment, including prophylaxis of inflammation enhanced cough in a mammal in need thereof.

Claims (269)

1. A compound of the formula:

wherein

R 1 is an optionally substituted aryl ring;

R 2 is independently selected from hydrogen, C 1-10 alkyl, C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, aryl, arylC 1-10 alkyl, heteroaryl, heteroarylC 1-10 alkyl, heterocyclic, or a heterocyclylC 1-10 alkyl moiety, which moieties, excluding hydrogen are all optionally substituted;

R 3 is an optionally substituted aryl;

R 10 is independently selected at each occurrence from hydrogen or C 1-4 alkyl;

X is (CH 2 ) n N(R 10 )S(O) m R 2 or (CH 2 ) n N(R 10 )C(O)R 2 ;

n is 0 or an integer having a value of 1 to 10;

m is 1 or 2;

or a pharmaceutically acceptable salt thereof.

2. The compound according to claim 1 which is Formula (I), or a pharmaceutically acceptable salt thereof.

3. The compound according to claim 1 which is Formula (Ia), or a pharmaceutically acceptable salt thereof.

4. The compound according to claim 1 wherein R 1 is an optionally substituted phenyl.

5. The compound according to claim 4 wherein the phenyl is substituted one or more times independently by halogen, alkyl, hydroxy, alkoxy, amino, or halosubstituted alkyl.

6. The compound according to claim 5 wherein the substituents are independently selected from fluorine, C 1-4 alkyl, or CF 3 .

7. The compound according to claim 5 wherein the phenyl ring is substituted in the 2, 4, or 6-position, di-substituted in the 2,4-position, or tri-substituted in the 2,4,6-position.

8. The compound according to claim 1 wherein X is (CH 2 ) n N(R 10 )S(O) m R 2 .

9. The compound according to claim 1 wherein X is (CH b 2 ) n N(R 10 )C(O)R 2 .

The compound according to claim 1 wherein R 2 is optionally substituted independently one or more times with C -10 alkyl, halo-substituted C 1-10 alkyl, C 2-10 alkenyl, C 2-10 alkynyl, C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, C 5-7 cycloalkenyl, C 5-7 cycloalkenyl C 1-10 alkyl, halogen, —CH(O), cyano, nitro, (CR 10 R 20 ) n OR 6 , (CR 10 R 20 ) n SH, (CR 10 R 20 ) n SR 7 , (CR 10 R 20 ) n S(O)R 7 , (CR 10 R 20 ) n S(O) 2 R 7 , (CR 10 R 20 ) n NR 10 S(O) 2 R 7 , (CR 10 R 20 ) n NR 4 R 14 , (CR 10 R 20 ) n CN, (CR 10 R 20 ) n S(O) 2 NR 4 R 14 , (CR 10 R 20 ) n C(Z)R 6 , (CR 10 R 20 ) n OC(Z)R 6 , (CR 10 R 20 ) n C(Z)OR 6 , (CR 10 R 20 ) n C(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10 C(Z)R 6 , (CR 10 R 20 ) n NR 10 C(═NR 10 )NR 4 R 14 , (CR 10 R 20 ) n C(═NOR 6 )NR 4 R 14 , (CR 10 R 20 ) n OC(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10 C(Z)NR 4 R 14 , or (CR 10 R 20 ) n NR 10 C(Z)OR 7 ; and wherein

R 4 and R 14 are each independently selected from hydrogen, optionally substituted C 1-4 alkyl, optionally substituted C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-4 alkyl, optionally substituted aryl, or optionally substituted aryl-C 1-4 alkyl, or R 4 and R 14 together with the nitrogen which they are attached form an optionally substituted heterocyclic ring of 4 to 7 members, which ring optionally contains an additional heteroatom selected from oxygen, sulfur or NR 9 ;

R 6 is hydrogen, C 1-10 alkyl, C 3-7 cycloalkyl, heterocyclyl, heterocyclyl C 1-10 alkyl, aryl, arylC 1-10 alkyl, heteroaryl or heteroarylC 1-10 alkyl, wherein each of these moieties may be optionally substituted;

R 9 is hydrogen, C(Z)R 6 or optionally substituted C 1-10 alkyl, optionally substituted aryl or optionally substituted aryl-C 1-4 alky;

R 7 is C 1-6 alkyl, aryl, arylC 1-6 alkyl, heterocyclic, heterocyclylC 1-6 alkyl, heteroaryl, or heteroarylC 1-6 alkyl; and wherein each of these moieties may be optionally substituted; and R 20 is independently selected at each occurrence from hydrogen or C 1-4 alky; and

Z is oxygen or sulfur.

11. The compound according to claim 9 wherein R 2 is an optionally substituted alkyl.

The compound according to claim 1 wherein R 3 is optionally substituted one or more times independently with C 1-10 alkyl, halo-substituted C 1-10 alkyl, C 2-10 alkenyl, C 2-10 alkynyl, C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, C 5-7 cycloalkenyl, C 5-7 cycloalkenylC 1-10 alkyl, halogen, cyano, nitro, (CR 10 R 20 ) n OR 6 , (CR 10 R 20 ) n SH, (CR 10 R 20 ) n SR 7 , (CR 10 R 20 ) n S(O)R 7 , (CR 10 R 20 ) n S(O) 2 R 7 , (CR 10 R 20 ) n NHS(O) 2 R 7 , (CR 10 R 20 ) n N 4 R 14 , (CR 10 R 20 ) n CN, (CR 10 R 20 ) n S(O) 2 NR 4 R , 14 (CR 10 R 20 ) n C(Z)R 6 , (CR 10 R 20 ) n OC(Z)R 6 , (CR 10 R 20 ) n C(Z)OR 6 , (CR 10 R 20 ) n C(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10 C(Z)R 6 , (CR 10 R 20 ) n NR 10 C(═NR 10 )NR 4 R 14 , (CR 10 R 20 ) n OC(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10C(Z)NR 4 R 14 , or (CR 10 R 20 ) n NR 10 C(Z)OR 7 ; and wherein

R 4 and R 14 are each independently selected from hydrogen, optionally substituted C 1-4 alkyl, optionally substituted C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-4 alkyl, optionally substituted aryl, or optionally substituted aryl-C 14 alkyl, or R 4 and R 14 together with the nitrogen which they are attached form an optionally substituted heterocyclic ring of 4 to 7 members, which ring optionally contains an additional heteroatom selected from oxygen, sulfur or NR 9 ;

R 6 is hydrogen, C 1-10 alkyl, C 3-7 cycloalkyl, heterocyclyl, heterocyclyl C 1-10 alkyl, aryl, arylC 1-10 alkyl, heteroaryl or heteroarylC 1-10 alkyl, wherein each of these moieties may be optionally substituted;

R 9 is hydrogen, C(Z)R 6 or optionally substituted C 1-10 alkyl, optionally substituted aryl or optionally substituted aryl-C 1-4 alky;

R 7 is C 1-6 alkyl, aryl, arylC 1-6 alkyl, heterocyclic, heterocyclylC 1-6 alkyl, heteroaryl, or heteroarylC 1-6 alkyl; and wherein each of these moieties may be optionally substituted; and R 20 is independently selected at each occurrence from hydrogen or C 1-4 alky; and

Z is oxygen or sulfur.

13. The compound according to claim 12 wherein the R 3 optional substituent is independently selected from halogen, alkyl, hydroxy, alkoxy, amino, or halosubstituted alkyl.

14. A compound which is:

2-Methylsulfanyl-4,8-diphenyl-8H-pyrido[2,3-d]pyrimidin-7-one;

2-Methanesulfonyl-4,8-diphenyl-8H-pyrido[2,3-d]pyrimidin-7-one;

2-(2-Diethylamino-ethylamino)-4,8-diphenyl-8H-pyrido[2,3-d]pyrimidin-7-one;

8-(2,6-Difluoro-phenyl)-4-(4-fluoro-2-methyl-phenyl)-2-methoxy-8H-pyrido[2,3-d]pyrimidin-7-one;

8-(2,6-Difluoro-phenyl)-2-ethoxy-4-(4-fluoro-2-methyl-phenyl)-8H-pyrido[2,3-d]pyrimidin-7-one;

2-Butoxy-8-(2,6-difluoro-phenyl)-4-(4-fluoro-2-methyl-phenyl)-8H-pyrido[2,3-d]pyrimidin-7-one;

8-(2,6-Difluoro-phenyl)-4-(4-fluoro-2-methyl-phenyl)-2-methylsulfanyl-5,8-dihydro-6H-pyrido[2,3-d]pyrimidin-7-one; or a pharmaceutically acceptable salt thereof.

15. A compound, which is:

8-(2,6-Difluoro-phenyl)-4-(4-fluoro-2-methyl-phenyl)-2-methylsulfanyl-8H-pyrido-[2,3-d]pyrimidin-7-one;

4,8-Bis-(2-fluoro-phenyl)-2-methylsulfanyl-8H-pyrido[2,3-d]pyrimidin-7-one;

8-(2-Chloro-phenyl)-4-(2-fluoro-phenyl)-2-methylsulfanyl-8H-pyrido-[2,3-d]pyrimidin-7-one;

4,8-Bis-(2-chloro-phenyl)-2-methylsulfanyl-8H-pyrido[2,3-d]pyrimidin-7-one;

18. The compound according to claim 17 , wherein the aryl ring of R 1 is optionally substituted, independently one or more times by halogen, C 1-4 alkyl, or halo-substituted-C 1-4 alkyl.

19. The compound according to claim 17 wherein the aryl ring of R 1 is a phenyl ring substituted in the 2, 4, or 6-position, di-substituted in the 2,4-position, or tri-substituted in the 2,4,6-position.

20. A compound of the formula:

R 4 and R 14 are each independently selected from hydrogen, optionally substituted C 1-4 alkyl, optionally substituted aryl, or optionally substituted aryl-C 1-4 alkyl, or R 4 and R 14 together with the nitrogen which they are attached form a heterocyclic ring of 5 to 7 members, which ring optionally contains an additional heteroatom selected from oxygen, sulfur or NR 9 ;

21. The compound according to claim 20 , wherein

R 2 is independently selected from C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, heterocyclic, or a heterocyclylC 1-10 alkyl moiety, which moieties are all optionally substituted; and

R 4 and R 14 together with the nitrogen which they are attached form a heterocyclic ring of 5 to 7 members, which ring optionally contains an additional heteroatom selected from oxygen, sulfur or NR 9 , wherein R 9 is hydrogen or optionally substituted C 1-10 alkyl, optionally substituted aryl or optionally substituted aryl-C 1-4 alkyl.

22. The compound according to claim 20 , wherein X is not hydrogen.

23. The compound according to claim 20 which is Formula (I), or a pharmaceutically acceptable salt thereof.

25. The compound according to claim 20 wherein R 1 is an optionally substituted phenyl.

26. The compound according to claim 25 wherein the phenyl is substituted one or more times independently by halogen, alkyl, hydroxy, alkoxy, amino, or halosubstituted alkyl.

27. The compound according to claim 26 wherein the substituents on the R 1 ring are independently selected from fluorine, C 1-4 alkyl, or CF 3 .

R 10 and R 20 are each independently selected from hydrogen or C 1-4 alkyl; R 11 is C 1-4 alkyl, halo-substituted C 1-4 alkyl, C 2-4 alkenyl, C 2-4 alkynyl, C 3-7 cycloalkyl, C 5-7 cycloalkenyl, aryl, arylC 1-4 alkyl, heteroaryl, heteroarylC 1-4 alkyl, heterocyclyl, heterocyclylC 1-4 alkyl, (CR 10 R 20 ) t OR 7 , (CR 10 R 20 ) t SR 7 , (CR 10 R 20 ) t S(O)R 7 , (CR 10 R 20 ) t S(O) 2 R 7 , (CR 10 R 20 ) t NHS(O) 2 R 7 , or (CR 10 R 20 ) v NR 4 R 14 ; and wherein the aryl, arylalkyl, heteroaryl, heteroarylalkyl may be optionally substituted;

t is an integer having a value of 1 to 3;

v is 0 or an integer having a value of 1 or 2;

R a′ is C 1-4 alkyl, halo-substituted C 1-4 alkyl, C 2-4 alkenyl, C 2-4 alkynyl, C 3-7 cycloalkyl, C 5-7 cycloalkenyl, aryl, arylC 1-4 alkyl, heteroaryl, heteroarylC 1-4 alkyl, heterocyclyl, heterocyclylC 1-4 alkyl, (CR 10 R 20 ) v OR 7 , (CR 10 R 20 ) v SR 7 , (CR 10 R 20 ) v S(O)R 7 , (CR 10 R 20 ) v S(O) 2 R 7 , (CR 10 R 20 ) v NHS(O) 2 R 7 ,or (CR 10 R 20 ) v NR 4 R 14 ; and wherein the aryl, arylalkyl, heteroaryl, heteroaryl alkyl may be optionally substituted.

29. The compound according to claim 28 wherein R 1 is an optionally substituted phenyl.

30. The compound according to claim 20 wherein X is OR 2 , or S(O) m R 2 .

31. The compound according to claim 20 wherein X is (CH2) n NR 4 R 14 , or (CH 2 ) n N(R 2 ) 2 .

32. The compound according to claim 20 wherein X is R 2 or S(O) m R 2 .

33. The compound according to claim 32 wherein X is R 2 and R 2 is an optionally substituted alkyl, aryl or arylalkyl.

34. The compound according to claim 30 wherein X is S(O) m R 2 and R 2 is an optionally substituted alkyl, aryl or arylalkyl.

35. The compound according to claim 34 wherein R 2 is an optionally substituted alkyl.

36. The compound according to claim 35 wherein R 2 is methyl.

37. R 10 and R 20 are each independently selected from hydrogen or C 1-4 alkyl.

38. A compound of the formula:

X is S(O) m R 2 ;

R 1 is an optionally substituted aryl;

R 2 is independently selected from C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, heterocyclic or heterocyclicC 1-10 alkyl;

R 3 is an optionally substituted aryl;

m is 1 or 2;

or a pharmaceutically acceptable salt thereof.

39. The compound according to claim 38 which is Formula (I), or a pharmaceutically acceptable salt thereof.

40. The compound according to claim 38 which is Formula (Ia), or a pharmaceutically acceptable salt thereof.

41. The compound according to claim 38 wherein R 1 is an optionally substituted phenyl or napthyl.

42. The compound according to claim 41 wherein the phenyl is substituted one or more times independently by halogen, alkyl, hydroxy, alkoxy, amino, or halosubstituted alkyl.

43. The compound according to claim 41 wherein the phenyl ring is substituted in the 2, 4, or 6-position, di-substituted in the 2,4- position, or tri-substituted in the 2,4,6-position, wherein X is S(O)R 2 .

44. The compound according to claim 38 wherein R 3 is optionally substituted one or more times independently with C 1-10 alkyl, halo-substituted C 1-10 alkyl, C 2-10 alkenyl, C 2-10 alkynyl, C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-10 alkyl, C 5-7 cycloalkenyl, C 5-7 cycloalkenyl C 1-10 alkyl, halogen, (CR 10 R 20 ) n OR 6 , (CR 10 R 20 ) n SH, (CR 10 R 20 ) n SR 7 (CR 10 R 20 ) n S(O)R 7 , (CR 10 R 20 ) n S(O) 2 R 7 , (CR 10 R 20 ) n NHS(O) 2 R 7 , (CR 10 R 20 ) n N 4 R 14 , (CR 10 R 20 ) n CN, (CR 10 R 20 ) n S(O) 2 NR 4 R 14 , (CR 10 R 20 ) n C(Z)R 6 , (CR 10 R 20 ) n OC(Z)R 6 , (CR 10 R 20 ) n C(Z)OR 6 , (CR 10 R 20 ) n C(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10 C(Z)R 6 , (CR 10 R 20 ) n NR 10 C(═NR 10 )NR 4 R 14 , (CR 10 R 20 ) n OC(Z)NR 4 R 14 , (CR 10 R 20 ) n NR 10 C(Z) NR 4 R 14 , or (CR 10 R 20 ) n NR 10C(Z)OR 7 ; and wherein

R 7 is C 1-6 alkyl, aryl, arylC 1-6 alkyl, heterocyclic, heterocyclylC 1-6 alkyl, heteroaryl, or heteroarylC 1-6 alkyl; and wherein each of these moieties may be optionally substituted;

R 4 and R 14 are each independently selected from hydrogen, optionally substituted C 1-4 alkyl, optionally substituted C 3-7 cycloalkyl, C 3-7 cycloalkylC 1-4 alkyl, optionally substituted aryl, or optionally substituted aryl-C 1-4 alkyl, or R 4 and R 14 together with the nitrogen which they are attached form an optionally substituted heterocyclic ring of 4 to 7 members, which ring optionally contains an additional heteroatom selected from oxygen, sulfur or NR 9 ;

R 9 is hydrogen, C(Z)R 6 or optionally substituted C 1-10 alkyl, optionally substituted aryl or optionally substituted aryl-C 1-4 alkyl;

R 10 and R 20 are each independently selected from hydrogen or C 1-4 alkyl;

n is 0 or an integer having a value of 1 to 10; and

Z is oxygen or sulfur.

45. The compound according to claim 44 wherein R 3 is an optionally substituted phenyl.

46. The compound according to claim 45 wherein the optional substituents on the R 3 phenyl are independently selected from halogen, alkyl, hydroxy, alkoxy, amino, or halosubstituted alkyl.

47. The compound according to claim 38 wherein R 3 is an optionally substituted phenyl.

48. A pharmaceutical composition comprising an effective amount of a compound according to claim 38 and a pharmaceutically acceptable carrier or diluent.

49. The compound according to claim 38 wherein m is 1 and X is S(O)R 2 .

50. The compound according to claim 49 wherein R 2 is methyl.

51. A pharmaceutical composition comprising an effective amount of a compound according to any one of claims 1 , 14 , 15 , or 16 and a pharmaceutically acceptable carrier or diluent.

52. A pharmaceutical composition comprising an effective amount of a compound according to claim 20 and a pharmaceutically acceptable carrier or diluent.
