Abstract

Compositions and methods are provided for potentiating the activity of the mitogen-activated protein kinase p38. In particular the mitogen-activated protein kinase kinase MEK6, and variants thereof that stimulate phosphorylation of p38 are provided. Such compounds may be used, for example, for therapy of diseases associated with the p38 cascade and to identify antibodies and other agents that inhibit or activate signal transduction via p38.

Claims (13)

1. An isolated polynucleotide encoding a polypeptide comprising:

(a) a polypeptide variant of TAO1 kinase, comprising an amino acid sequence that comprises residues 1–416, 1–320, or 15–285 of SEQ ID NO:2, wherein the variant retains an ability to phosphorylate MEK3 that is at least 90% of the ability of a native TAO1 kinase to phosphorylate MEK3 under the same conditions, with the proviso that the variant does not comprise more than 500 consecutive amino acids of SEQ ID NO:2; or

(b) a polypeptide variant of TAO2 kinase, comprising an amino acid sequence that comprises residues 1–416, 1–320, or 15–285 of SEQ ID NO:4, wherein the variant active retains an ability to phosphorylate MEK3 that is at least 90% of the ability of a native TAO2 kinase to phosphorylate MEK3 under the same conditions, with the proviso that the variant does not comprise more than 500 consecutive amino acids of SEQ ID NO:4.

2. The isolated polynucleotide according to claim 1 , wherein the polynucleotide comprises at least 800 consecutive nucleotides of SEQ ID NO: 1.

3. The isolated polynucleotide according to claim 1 , wherein the polynucleotide comprises at least 800 consecutive nucleotides of SEQ ID NO:3.

4. A recombinant expression vector comprising a polynucleotide according to claim 1 .

5. A host cell transformed or transfected wit an expression vector according to claim 4 .

6. A composition, comprising:

(a) a polynucleotide according to claim 1 ; and

(b) a physiologically acceptable carrier.

The isolated polynucleotide of claim 1 , wherein the polynucleotide encodes a polypeptide comprising a polypeptide variant of TAO1 kinase, comprising an amino acid sequence that comprises residues 1–416, 1–320, or 15–285 of SEQ ID NO:2, wherein the variant retains an ability to phosphorylate MEK3 that is at least 90% of the ability of a native TAO1 kinase to phosphorylate MEK3 under the same conditions, with the proviso that the variant does not comprise more than 500 consecutive amino acids of SEQ ID NO:2.

The isolated polynucleotide of claim 1 , wherein the polynucleotide encodes a polypeptide comprising a polypeptide variant of TAO2 kinase, comprising an amino acid sequence that comprises residues 1–416, 1–320, or 15–285 of SEQ ID NO:4, wherein the variant retains an ability to phosphorylate MEK3 that is at least 90% of the ability of a native TAO2 kinase to phosphorylate MEK3 under the same conditions, with the proviso that the variant does not comprise more than 500 consecutive amino acids of SEQ ID NO:4.

An isolated polynucleotide encoding a polypeptide comprising a polypeptide variant of TAO2 kinase, comprising an amino acid sequence that is at least 90% identical to residues 15–285 of SEQ ID NO:4, wherein the variant retains an ability to phosphorylate MEK3 that is at least 90% of the ability of a native TAO2 kinase to phosphorylate MEK3 under the same conditions, with the proviso that the variant does not comprise more than 500 consecutive amino acids of SEQ ID NO:4.
