Abstract

The invention provides, among other aspects, compositions and methods for treating, preventing, and diagnosing diseases or conditions associated with an abnormal level or activity of biglycan; diseases or conditions associated with an abnormal level or activity of collagen VI; disorders associated with an unstable cytoplasmic membrane, due, e.g., to an unstable dystrophin associated protein complex (DAPC); and disorders associated with abnormal synapses or neuromuscular junctions, including those resulting from an abnormal MuSK activation or acetylcholine receptor (AChR) aggregation.

Claims (12)

1. A method for treating a deficiency in collagen VI expression levels in mammalian muscle, comprising administering to a mammal a biglycan polypeptide in an amount sufficient to increase collagen VI expression levels in the mammalian muscle, wherein the biglycan polypeptide comprises an amino acid sequence which is at least about 95% identical to amino acids 38-368 of SEQ ID NO: 9, or a portion thereof, and which has a biological activity of human biglycan.

2. The method of claim 1 , wherein the biglycan polypeptide binds to MuSK.

3. The method of claim 1 , wherein the biglycan polypeptide binds to a α-sarcoglycan and/or γ-sarcoglycan.

4. The method of claim 1 , wherein the biglycan polypeptide binds to a collagen VI polypeptide.

5. The method of claim 1 , wherein the biglycan polypeptide induces phosphorylation of sarcoglycans.

6. The method of claim 1 , wherein the biglycan polypeptide upregulates utrophin levels.

7. The method of claim 1 , wherein the mammal has a condition selected from the group consisting of Bethlem myopathy, and Ullrich Congenital Muscular Dystrophy.

8. The method of claim 1 , wherein the biglycan polypeptide comprises amino acids 38-368 of SEQ ID NO: 9.

9. The method of claim 1 , wherein the biglycan polypeptide comprises amino acids 20-368 of SEQ ID NO: 9.

10. The method of claim 1 , wherein the biglycan polypeptide comprises at least one repeat motif of 24 amino acids in the Leucine Rich Repeat (LRR) of SEQ ID NO: 9.

11. The method of claim 1 , wherein the biglycan polypeptide is encoded by a nucleic acid which hybridizes under conditions of 6.0+sodium chloride/sodium citrate (SSC) at about 45° C. to a complementary strand of SEQ ID NO: 8.

12. The method of claim 1 , wherein the biglycan polypeptide comprises the amino acid sequence of SEQ ID NO: 9.
