NIMA interacting proteins
View Patent ↗A novel class of NIMA interacting proteins (PIN), exemplified by Pin1, is provided. Pin1 induces a G2 arrest and delays NIMA-induced mitosis when overexpressed, and triggers mitotic arrest and DNA fragmentation when depleted. Methods of identifying other Pin proteins and Pin-interacting proteins and identifying compositions which affect Pin activity or expression are also provided.
1. A method for determining whether a composition inhibits the peptidyl-prolyl isomerase activity of a Pin1 protein having the amino acid sequence set forth in SEQ ID NO: 2, said method comprising:
incubating the composition with a Pin1 protein having the amino acid sequence set forth in SEQ ID NO: 2 or a functional fragment thereof, or with a recombinant cell expressing the Pin1 protein or a functional fragment thereof, under conditions sufficient to allow the composition to interact with the Pin1 protein or functional fragment thereof; wherein the functional fragment of the Pin1 protein has peptidyl prolyl isomerase activity; and
determining the effect of the composition on the peptidyl-prolyl isomerase activity of the Pin1 protein.
2. The method of claim 1 , wherein the functional fragment comprises at least amino acid residues 59–163 of SEQ ID NO: 2.
3. The method of claim 1 , wherein the peptidyl prolyl isomerase activity is not inhibited by cyclosporine A or FK520.
