Streptococcal C5a peptidase vaccine
Novel vaccines for use against β-hemolytic Streptococcus colonization or infection are disclosed. The vaccines contain an immunogenic amount of a variant of streptococcal C5a peptidase (SCP). Also disclosed is a method of protecting a susceptible mammal against β-hemolytic Streptococcus colonization or infection by administering such a vaccine. Enzymatically inactive SCP, and polynucleotides encoding these SCP proteins are further disclosed.
1. A composition comprising an isolated enzymatically inactive streptococcal C5a peptidase (SCP) conjugated or linked to a polysaccharide, wherein the SCP comprises the specificity crevice or catalytic domain of SCP.
2. The composition of claim 1 , wherein the polysaccharide is a group A streptococcal polysaccharide, a C-polysaccharide from group B Streptococci , or a capsular polysaccharide of S. pneumoniae or group B Streptococci .
3. The composition of claim 1 , wherein the polysaccharide is a polysaccharide of a pathogen.
4. The composition of claim 1 , wherein the polysaccharide is a Streptococcus pneumoniae capsular polysaccharide.
5. The composition of claim 1 , wherein the SCP comprises the catalytic domain of SCP.
6. The composition of claim 5 , wherein the SCP further comprises the region of SCP between the catalytic domain and cell wall insert.
7. The composition of claim 1 , wherein the SCP comprises the specificity crevice.
8. The composition of claim 1 , wherein the SCP does not contain the SCP signal sequence of said SCP.
9. The composition of claim 1 , wherein the SCP does not contain the SCP cell wall insert of said SCP.
10. The composition of claim 1 , wherein the SCP has a modification at one or more of its amino acid residues 260, 261, 262, 415, 416, 417, 130, 193, 295 or 512.
