Abstract

The present invention provides anti-inflammatory compounds, pharmaceutical compositions thereof, and methods of use thereof for treating inflammatory disorders. The present invention also provides methods of identifying anti-inflammatory compounds and methods of inhibiting NF-κB-dependent target gene expression in a cell.

Claims (10)

1. An anti-inflammatory compound comprising an NE-κB Essential Modulator (NEMO) binding domain fused with at least one cell membrane translocation domain, wherein said NEMO binding domain comprises an amino acid sequence selected from the group consisting of SEQ ID NO:2, SEQ ID NO:4, SEQ ID NO:5, SEQ ID NO:6, SEQ ID NO:8, SEQ ID NO:9, SEQ ID NO:11, SEQ ID NO:12, SEQ ID NO:14, SEQ ID NO: 16 and SEQ ID NO:17, and wherein said compound is less than one-hundred amino acids in length.

2. The anti-inflammatory compound of claim 1 , wherein the cell membrane translocation domain facilitates membrane translocation in vivo.

3. The anti-inflammatory compound of claim 1 , wherein the cell membrane translocation domain is selected from the group consisting of the third helix of the antennapedia homeodomain and HIV-1 Tat protein.

4. A composition comprising the anti-inflammatory compound of any one of claims 1 , 2 , or 3 .

5. The composition of claim 4 , further comprising a pharmaceutically acceptable carrier.

6. An anti-inflammatory compound comprising the amino acid sequence DRQIKIWFQNRRMKWKKTALDWSWLQTE (SEQ ID NO: 18).

7. A method of treating an NF-κB-mediated condition in a subject, comprising administering to the subject an effective amount of the anti-inflammatory compound of claim 1 .

8. The method of claim 7 , wherein the NF-κB-mediated condition is a condition selected from the group consisting of an inflammation disorder, an autoimmune disease, transplant rejection, osteoporosis, cancer, Alzheimer's disease, atherosclerosis, a viral infection, and ataxia telangiectasia.

The method of claim 8 , wherein the inflammation disorder is selected from the group consisting of asthma, allergies, uticaria, anaphylaxis, cutaneous inflammation, sepsis, psoriasis, rheumatoid arthritis, osteoarthritis, psoriatic arthritis, inflammatory bowel disease, chronic obstructive pulmonary disease, vasculitis, and bursitis.

10. The method of claim 8 , wherein the inflammation disorder is selected from the group consisting of dermititis, eczema, psoriasis, osteoarthritis, psoriatic arthritis, lupus, and spondylarthritis.
