Neuregulin based methods and compositions for treating cardiovascular diseases
View Patent ↗The present invention relates to compositions and methods for preventing, treating or delaying various cardiovascular diseases or disorders in mammals, particularly in humans. More particularly, the present invention provides for compositions and methods for preventing, treating or delaying various cardiovascular diseases or disorders using, inter alia, a neuregulin protein, or a functional fragment thereof, or a nucleic acid encoding a neuregulin protein, or a functional fragment thereof, or an agent that enhances production and/or function of said neuregulin.
1. A method for treating viral myocarditis or dilated (congestive) cardiomyopathy (DCM), in a mammal in need thereof, which method comprises administering to the mammal a neuregulin 1 protein in an amount from about 10 U/kg to about 400 U/kg of body weight of the mammal once a day for a period of between 3 days and 21 days.
2. The method of claim 1 , wherein the neuregulin 1 protein carries out its activity via binding with ErbB2-ErbB4 receptors.
3. The method of claim 1 , wherein the neuregulin 1 protein is neuregulin α2 or neuregulin β2.
4. The method of claim 1 , wherein the neuregulin 1 protein comprises the amino acid sequence of SEQ ID NO: 4.
5. The method of claim 1 , wherein the mammal is a human.
6. The method of claim 1 , wherein the viral myocarditis is caused by or associated with infection of a virus selected from the group consisting of Coxsackie Group A virus, Coxsackie Group B virus, ECHO virus and polio virus.
7. The method of claim 1 , wherein the viral myocarditis is caused by or associated with infection of a Coxsackie Group B virus.
8. The method of claim 1 , wherein the viral myocarditis is complicated by pericarditis or endocarditis.
9. The method of claim 1 , wherein the viral myocarditis has a clinical feature selected from the group consisting of myocardial damage, heart dysfunction, arrhythmia, systemic symptom and cardiomyopathy.
10. The method of claim 1 , wherein the viral myocarditis is an acute or a chronic viral myocarditis.
11. The method of claim 10 , wherein the acute viral myocarditis has a clinical feature selected from the group consisting of arrhythmia, heart failure and cardiac shock.
12. The method of claim 1 , wherein the viral myocarditis is prolonged with cardiac hypertrophy and/or permanent cardiac muscle injury.
13. The method of claim 1 , wherein the DCM has a clinical feature selected from the group consisting of ventricular hypertrophy, myocardial pump function failure and congestive heart failure.
14. The method of claim 1 , wherein the neuregulin 1 protein is administered with a pharmaceutically acceptable carrier or excipient.
15. The method of claim 1 , further comprising administering a prophylactic or therapeutic agent for viral myocarditis or DCM.
16. The method of claim 15 , wherein the neuregulin 1 protein is administered prior to, concurrently with, or subsequent to the administration of the prophylactic or therapeutic agent for viral myocarditis or DCM.
17. The method of claim 15 , wherein the prophylactic or therapeutic agent for viral myocarditis is selected from the group consisting of an antibiotic, a heart protective agent, an antioxidant and a nutrient for myocardium.
18. The method of claim 17 , wherein the antibiotic is penicillin.
19. The method of claim 17 , wherein the heart protective agent is taurine.
20. The method of claim 17 , wherein the antioxidant is selected from the group consisting of vitamin C, vitamin E and coenzyme Q 10.
21. The method of claim 17 , wherein the nutrient for myocardium is an energy combination.
22. The method of claim 15 , wherein the prophylactic or therapeutic agent for DCM is selected from the group consisting of a cardiac tonic, a diuretic, an angiotensin 1-converting enzyme inhibitor (ACEI), a calcium antagonist and a β-receptor antagonist.
23. The method of claim 22 , wherein the cardiac tonic is digoxin or cedilanid.
24. The method of claim 22 , wherein the diuretic is selected from the group consisting of an inhibitor of carbonic anhydrase, an osmotic diuretic, an inhibitor of Na + —K+—2Cl − symport, an inhibitor of Na + —Cl − symport, an inhibitor of renal epithelial Na+channels and an antagonist of mineralocorticoid receptors.
25. The method of claim 22 , wherein the calcium antagonist is amlodipine.
26. The method of claim 22 , wherein the β-receptor antagonist is carvedilol.
