Abstract

The present invention provides αVβ3 integrin cysteine loop domain agonists and antagonists (including peptide agonists and antagonists and analogs thereof), along with methods of using the same.

Claims (17)

1. An isolated antibody that: (a) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a human β3 integrin, wherein said amino acids consist of SEQ ID NO: 82; (b) does not specifically bind to the RGD binding site of a human β3 integrin or utilize the RGD binding site of human β3 integrin; and (c) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a pig β3 integrin, wherein said amino acids consist of SEQ ID NO: 82.

2. The isolated antibody of claim 1 , coupled to a detectable group.

3. The isolated antibody of claim 1 , coupled to a therapeutic group.

4. A monoclonal antibody that: (a) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a human β3 integrin, wherein said amino acids consist of SEQ ID NO: 82; (b) does not specifically bind to the RGD binding site of a human β3 integrin or utilize the RGD binding site of the human β3 integrin; and (c) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a pig β3 integrin, wherein said amino acids consist of SEQ ID NO: 82.

5. The antibody of claim 4 , coupled to a detectable group.

6. The antibody of claim 4 , coupled to a therapeutic group.

7. The isolated antibody of claim 1 , wherein said antibody is purified.

8. The isolated antibody of claim 1 , wherein said antibody does not specifically inhibit ligand occupancy of the RGD binding site of the β3 integrin.

9. The antibody of claim 4 , wherein said antibody does not specifically inhibit ligand occupancy of the RGD binding site of the β3 integrin.

10. An isolated antibody that: (a) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a human β3 integrin, wherein said amino acids consist of SEQ ID NO: 82; (b) does not specifically bind to the RGD binding site of a human β3 integrin or stimulate the specific biochemical events that are stimulated by peptides that bind to the RGD binding site of human β3 integrin; and (c) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a pig β3 integrin, wherein said amino acids consist of SEQ ID NO: 82.

A monoclonal antibody that: (a) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a human β3 integrin, wherein said amino acids consist of SEQ ID NO: 82; (b) does not specifically bind to the RGD binding site of a human β3 integrin or stimulate the specific biochemical events that are stimulated by peptides that bind to the RGD binding site of human β3 integrin; and (c) specifically binds to the cysteine loop domain at amino acids 177 to 184 of a pig β3 integrin, wherein said amino acids consist of SEQ ID NO: 82.

12. The isolated antibody of claim 10 , coupled to a detectable group.

13. The isolated antibody of claim 10 , coupled to a therapeutic group.

14. The antibody of claim 11 , coupled to a detectable group.

15. The antibody of claim 11 , coupled to a therapeutic group.

16. The isolated antibody of claim 10 , wherein said antibody does not specifically inhibit ligand occupancy of the RGD binding site of the β3 integrin.

17. The antibody of claim 11 , wherein said antibody does not specifically inhibit ligand occupancy of the RGD binding site of the β3 integrin.
