Abstract

The present invention provides porcine Forssman synthetase (FSM synthase) (Globoside α-N-acetylgalactosaminyltransferase) protein, cDNA, and genomic DNA sequence. Furthermore, the present invention includes porcine animals, tissue and organs as well as cells and cell lines derived from such animals, tissue and organs, which lack expression of functional FSM synthetase. Such animals, tissues, organs and cells can be used in research and in medical therapy, including in xenotransplantation. In addition, methods are provided to prepare organs, tissues, and cells lacking the porcine FSM synthetase gene for use in xenotransplantation.

Claims (7)

1. An isolated nucleic acid comprising at least 1,350 contiguous nucleotides of SEQ ID NO: 16 wherein the nucleic acid can be used to functionally inactivate a Forssman synthetase gene in a cell.

2. A nucleic acid construct comprising at least 1,350 contiguous nucleotides of SEQ ID NO: 16 wherein the construct can be used to functionally inactivate a Forssman synthetase gene in a cell.

3. The construct of claim 2 wherein the construct is a vector.

4. The construct of claim 2 wherein the construct is a plasmid.

5. The construct of claim 2 wherein the construct further includes a marker gene.

6. An isolated host genetically modified cell transformed with the nucleic acid of claim 1 .

7. The cell of claim 6 wherein the cell is a porcine cell.
