Abstract

The invention relates to isolated insecticidal polypeptides expressed in the venom gland of spiders of the genera Atrax and Hadronyche have been described. Methods of treating insects, insect larvae, and plants with the insecticidal polypeptides are also described.

Claims (15)

1. A purified polypeptide comprising an amino acid sequence of any one of SEQIDNO: 1, 2, 5 and 6.

2. The purified polypeptide of claim 1 comprising SEQ ID NO: 2 and 6 said polypeptide further comprising a propeptide sequence, a signal peptide sequence, or a combination thereof.

3. A purified polypeptide having at least 85% sequence identity to SEQ ID NO: 2, wherein the polypeptide has insecticidal activity and having a molecular mass of approximately 4,300 Daltons.

4. An insecticidal composition comprising an insecticidally effective amount of the purified polypeptide of claim 3 , and an agriculturally acceptable carrier.

A method of controlling an insect or an insect larva, comprising contacting the insect or insect larva with an insecticidally effective amount of a U-ACTX polypeptide, wherein the U-ACTX polypeptide comprises the amino acid sequence of SEQ ID NO: 2, or 6 wherein the U-ACTX polypeptide has insecticidal activity.

6. The method of claim 5 , wherein the U-ACTX polypeptide comprises SEQ ID NO: 2.

7. The method of claim 5 , wherein the U-ACTX polypeptide is in the form of a purified polypeptide.

8. The method of claim 5 , wherein the U-ACTX polypeptide is impregnated in a collar.

9. A method of treating a plant, comprising contacting the plant with an insecticidally effective amount of a U-ACTX polypeptide, wherein the U- ACTX polypeptide comprises an amino acid sequence having a molecular mass of approximately 4,300 Daltons that is at least 85% identical to SEQ ID NO: 2, wherein the U-ACTX polypeptide has insecticidal activity.

10. The method of claim 9 , wherein the U-ACTX polypeptide is in the form of a purified polypeptide.

11. An insecticidal polypeptide, comprising an amino acid sequence that is at least 85% identical to SEQ ID Nos: 1, 2, 5,6, 11 and 12 and having a molecular mass of approximately 4,300 Daltons that blocks at least 50% of a calcium current in an insect voltage-gated calcium channel and at least 50% of activity of an insect high conductance calcium-activated potassium channel.

12. The insecticidal polypeptide of claim 11 , wherein the polypeptide comprises three intrachain disulfide bonds.

13. The insecticidal polypeptide of claim 11 , wherein the polypeptide is a component of the venom of a spider of the genera Atrax of Hadronyche.

14. The insecticidal polypeptide of claim 11 , wherein the insect voltage-gated calcium channel is one from a DUM nuron or P. americana.

15. The insecticidal polypeptide of claim 11 , wherein the high conductance insect calcium-activated potassium channel comprises a P. americana high conductance calcium-activated potassium channel.
