Abstract

An isolated, biologically active 33 kDa Hemagglutinin purified from the type A Clostridium botulinum neurotoxin complex and its uses are described.

Claims (8)

1. A method of eliciting an immune response against a clostridial neurotoxin in an animal, the method comprising administering to the animal a composition consisting essentially of a purified, biologically active hemagglutinin-33 polypeptide (Hn-33) from type A Clostridium botulinum, an isolated and purified clostridial neurotoxin polypeptide that inhibits neurotransmitter release from a presynaptic membrane of a neurosensory junction and has protease activity, and optionally a pharmaceutical carrier suitable for administration to an animal, wherein the Hn-33 polypeptide enhances the protease activity of the clostridial neurotoxin polypeptide.

2. The method of claim 1 , wherein the composition further comprises an adjuvant.

3. The method of claim 1 , wherein the neurotoxin is selected from the group consisting of type A botulinum neurotoxin, type E botulinum neurotoxin, and tetanus toxin.

4. The method of claim 1 , wherein the neurotoxin is type A botulinum neurotoxin.

5. The method of claim 1 , wherein the composition is administered orally to the animal.

6. The method of claim 1 , wherein the composition further comprises a biodegradable polymer that enables slow release of the neurotoxin.

7. The method of claim 1 , wherein the animal is a human.

8. A method of eliciting an immune response against a neurotoxin in an animal, the method comprising administering to the animal a composition comprising a purified, biologically active hemagglutinin-33 polypeptide (Hn-33) from type A Clostridium botulinum and a purified clostridial neurotoxin polypeptide, wherein the Hn-33 is protease-resistant and enhances the protease activity of said clostridial neurotoxin.
