Abstract

The present invention relates to the identification of a gene, now designated negevin (ngvn), that is involved in the genetic disease Bardet Biedl Syndrome (BBS), which is characterized by such diverse symptoms as obesity, diabetes, hypertension, mental retardation, renal cancer and other abnormalities, retinopathy and hypogonadism. The human NGVN protein disclosed herein is 731 amino acids in length and is coded for by a gene spanning 17 exons. Homologs have been identified in mouse, rat, zebrafish. Methods of use for the gene, for example in diagnosis and therapy of BBS and in drug screening, also are described.

Claims (12)

1. An isolated and purified nucleic acid encoding a human negevin (NGVN) polypeptide variant consisting of SEQ ID NO:2 but having one, two, three, four, five or all six of the following changes: Val75Gly, Arg272Stp, Arg275Stp, Ile123Val, or a frameshift starting at residue 314 or residue 403 of SEQ ID NO:2.

2. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the Val75Gly variant.

3. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the Arg272Stp variant.

4. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the Arg275Stp variant.

5. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the Ile123Val variant.

6. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the frameshift starting at residue 314.

7. The isolated and purified nucleic acid of claim 1 , wherein the nucleic acid comprises the sequence of SEQ ID NO:2 and consisting of the frameshift starting at residue 403.

A method of expressing a NGVN polypeptide variant comprising transforming a host cell with an expression construct encoding a NGVN polypeptide variant consisting of SEQ ID NO:2 but having one, two, three, four, five or all six of the following changes: Val75Gly, Arg272Stp, Arg275Stp, Ile123Val, or a frameshift starting at residue 314 or residue 403 of SEQ ID NO:2 and culturing said host cell under conditions supporting expression of said NGVN polypeptide variant.

9. The method of claim 8 , wherein said host cell is a prokaryotic cell.

10. The method of claim 8 , wherein said host cell is a eukaryotic cell.

11. The method of claim 8 , further comprising purifying said NGVN polypeptide variant.

12. The method of claim 8 , wherein said expression construct comprises an inducible promoter, and said method further comprises providing to said host cell and inducer of said promoter.
