Abstract

The present invention relates to a disintegrin and metalloproteinase containing thrombospondin 1-like domains (ADAMTS) and in particular to a novel ADAMTS13 protease and to nucleic acids encoding ADAMTS13 proteases. The present invention encompasses both native and recombinant wild-type forms of ADAMTS13, as well as mutant and variant forms including fragments, some of which posses altered characteristics relative to the wild-type ADAMTS13. The present invention also relates to methods of using ADAMTS13, including for treatment of TTP. The present invention also relates to methods for screening for the presence of TTP. The present invention further relates to methods for developing anticoagulant drugs based upon ADAMTS13.

Claims (12)

1. An isolated nucleic acid sequence having the nucleic acid sequence of SEQ ID NO: 1.

2. An isolated expression vector comprising the isolated nucleic acid sequence of claim 1 .

3. The isolated expression vector according to claim 2 , wherein the expression vector is a bacterial vector.

4. The isolated expression vector according to claim 3 , wherein the vector is selected from the group consisting of pQE70, pQE60, pQE-9, pBS, pD10, phagescript, psiX174, pbluescript SK, pBSKS, pNH8A, pNH16a, pNH18A, pNH46A, ptrc99a, pKK223-3, pKK233-3, pDR540, and pRIT5.

5. The isolated expression vector according to claim 1 , wherein the expression vector is a eukaryotic vector.

6. The isolated expression vector according to claim 5 , wherein the vector is selected from the group consisting of pWLNEO, pSV2CAT, pOG44, PXT1, pSG, pSVK3, pBPV, pMSG, pSVL.

7. The isolated expression vector according to claim 1 , wherein the expression vector is a baculovirus vector.

8. The isolated expression vector according to claim 7 , wherein the vector is selected from the group consisting of pPbac and pMbac.

9. An isolated host cell comprising an expression vector of claim 1 .

10. The isolated host cell of claim 9 , wherein the host cell is a eukaryotic cell.

11. The isolated host cell of claim 10 , wherein the eukaryotic cell is selected from the group consisting of a mammalian cell, an insect cell and a yeast cell.

12. The isolated host cell of claim 10 , wherein the eukaryotic cell is selected from the group consisting of a C127 cell, 3T3 cell, 293 cell, 293T cell, HeLa cell, BHK cell and a Chinese-hamster ovary (CHO) cell.
