Abstract

The present invention is directed to isolated polypeptides and antibodies suitable for producing therapeutic preparations, methods, and kits relating to bone deposition. One objective of the present invention is to provide compositions that improve bone deposition. Yet another objective of the present invention is to provide methods and compositions to be utilized in diagnosing bone dysregulation. The therapeutic compositions and methods of the present invention are related to the regulation of Wise, Sost, and closely related sequences. In particular, the nucleic acid sequences and polypeptides include Wise and Sost as well as a family of molecules that express a cysteine knot polypeptide.

Claims (10)

1. An isolated monoclonal antibody that specifically binds to the amino acid sequence depicted in SEQ ID NO: 46.

2. The isolated monoclonal antibody of claim 1 , wherein the antibody is a humanized antibody.

3. An isolated monoclonal antibody that specifically binds to the amino acid sequence is depicted in SEQ ID NO: 67.

4. The isolated monoclonal antibody of claim 3 , wherein the antibody is a humanized antibody.

5. An isolated monoclonal antibody that specifically binds to a Sost peptide depicted in SEQ ID NO: 46.

6. The isolated monoclonal antibody of claim 5 , wherein the antibody is a humanized antibody.

7. An isolated monoclonal antibody that specifically binds to a Sost peptide depicted in SEQ ID NO: 67.

8. The isolated monoclonal antibody of claim 7 , wherein the antibody is a humanized antibody.

9. An isolated monoclonal antibody that binds to the amino acid sequence depicted in SEQ ID NO: 46, wherein the isolated monoclonal antibody binds to the amino acid sequence with a specific binding activity (K a ) of at least about 10 7 mol −1 or greater.

10. An isolated monoclonal antibody that binds to an the amino acid sequence depicted in SEQ ID NO: 67, wherein the isolated monoclonal antibody binds to the amino acid sequence with a specific binding activity (K a ) of at least about 10 7 mol −1 or greater.
