Abstract

Compositions and methods for enhancing the immune response of a mammal to an antigen by engaging the OX-40 receptor on the surface of T-cells are disclosed, comprising administering to the mammal a composition comprising a purified OX-40 receptor binding agent and a pharmaceutically acceptable carrier, wherein the composition is administered to the mammal such that the OX-40 receptor binding agent is presented to T-cells of the mammal during or shortly after priming of the T-cells by the antigen. Such compositions and methods can be used in immunization and cancer treatment.

Claims (11)

1. A method of enhancing the immune response of a mammal to an antigen, comprising administering to the mammal a composition comprising a purified OX-40-ligand (OX-40L) and a pharmaceutically acceptable carrier, wherein said composition is administered to the mammal such that the OX-40L is presented to T-cells of the mammal during or shortly after priming of the T-cells by the antigen, wherein the composition comprises about 10 ng to 1 gram of OX-40L.

2. A method according to claim 1 wherein the OX-40L is administered to the mammal about 3-7 days after administration of the antigen.

3. A method according to claim 1 wherein the antigen is selected from the group consisting of viral antigens, bacterial antigens and tumor antigens.

4. A method for stimulating the immune response of a mammal to a tumor cell in the mammal, comprising administering to the mammal a therapeutically effective dose of a purified OX-40 ligand (OX-40L).

5. A method of causing enhanced immune response against a tumor in a mammal, the method comprising increasing the amount of an X-40 ligand (OX-40L) at the tumor site.

6. The method of claim 1 , wherein the composition comprises about 10 μg to 100 mg of OX-40L.

7. The method of claim 1 , wherein the composition comprises 1 mg or 10 mg of OX-40L.

8. The method of claim 1 , wherein the OX-40L is administered intravenously.

9. The method of claim 3 , wherein the antigen is a bacterial antigen.

10. The method of claim 9 , wherein the bacterial antigen is a vaccine component.

11. The method of claim 5 , wherein the tumor is a breast carcinoma, a lung carcinoma, a pancreas carcinoma, an ovary carcinoma, a kidney carcinoma, a colon carcinoma, a bladder carcinoma, melanoma or a sarcoma.
