Abstract

The present invention relates to oncogenes or tumor suppressor genes, as well as other genes, involved in prostate cancer and their expression products, as well as derivatives and analogs thereof. Provided are therapeutic compositions and methods of detecting and treating cancer, including prostate and other related cancers. Also provided are methods of diagnosing and/or prognosing prostate cancer by determining the expression level of at least one prostate cancer-cell-specific gene, including, for example, the ERG gene or the LTF gene alone, or in combination with at least one of the AMACR gene and the DD3 gene.

Claims (20)

1. A kit for detecting prostate cancer, comprising oligonucleotide probes, wherein the oligonucleotide probes consist of a first oligonucleotide probe capable of hybridizing to a first overexpressed prostate cancer cell specific gene comprising the sequence of SEQ ID NO: 1 or a sequence complementary thereto under conditions of high stringency and a second oligonucleotide probe capable of hybridizing to a second overexpressed prostate cancer cell specific gene comprising the sequence of SEQ ID NO: 3 or a sequence complementary thereto under conditions of high stringency, wherein the oligonucleotide probes are labeled with a detectable group, wherein the high stringency conditions comprise hybridization for 48 hours at 65° C. in 6×SSC followed by a wash in 0.1×SSC at 50° C. for 45 minutes.

A kit for detecting prostate cancer, comprising oligonucleotide probes, wherein the oligonucleotide probes consist of a first oligonucleotide probe capable of hybridizing to a first overexpressed prostate cancer cell specific gene comprising the sequence of SEQ ID NO: 1 or a sequence complementary thereto under conditions of high stringency and a second overexpressed oligonucleotide probe capable of hybridizing to a second prostate cancer cell specific gene comprising the sequence of SEQ ID NO:4 or a sequence complementary thereto under conditions of high stringency, wherein the oligonucleotide probes are labeled with a detectable group, wherein the high stringency conditions comprise hybridization for 48 hours at 65° C. in 6×SSC followed by a wash in 0.1×SSC at 50° C. for 45 minutes.

3. The kit of claim 2 , wherein the first and second oligonucleotide probes are fixed to a support.

4. The kit of claim 1 or 2 , wherein the first oligonucleotide probe comprises at least about 20 contiguous nucleotides of SEQ ID NO: 1, or a sequence complementary thereto.

The kit of claim 1 or 2 , further comprising a first oligonucleotide primer and a second oligonucleotide primer, wherein the first oligonucleotide primer contains a sequence that hybridizes to a first sequence in SEQ ID NO:1, and the second oligonucleotide primer contains a sequence that hybridizes to a second sequence in a nucleic acid strand complementary to SEQ ID NO:1, wherein the first sequence does not overlap with the second sequence, and wherein the first and second oligonucleotide primers are capable of amplifying a target sequence in SEQ ID NO:1.

6. The kit of claim 5 , wherein the first oligonucleotide primer comprises at least about 17 nucleotides and wherein the second oligonucleotide primer comprises at least about 17 nucleotides.

7. The kit of claim 3 , wherein the solid support is a micron- or submicron-sized bead or sphere.

A kit for detecting prostate cancer, comprising: labeled oligonucleotide probes, wherein the labeled oligonucleotide probes comprise: a first labeled oligonucleotide probe comprising at least 15 contiguous nucleotides of nucleotides 510-773 of SEQ ID NO: 1 or a sequence complementary thereto and a second labeled oligonucleotide probe comprising at least 15 contiguous nucleotides of SEQ ID NO: 4 or a sequence complementary thereto.

A kit for detecting prostate cancer, comprising oligonucleotide probes, wherein the oligonucleotide probes consist of one or more first oligonucleotide probes comprising at least 15 contiguous nucleotides of SEQ ID NO: 1 or a sequence complementary thereto and one or more second oligonucleotide probes comprising at least 15 contiguous nucleotides of SEQ ID NO: 4 or a sequence complementary thereto, wherein the one or more first and second oligonucleotide probes are labeled with at least one detectable group.

10. The kit of claim 9 , wherein the one or more first oligonucleotide probes comprise at least 20 contiguous nucleotides of SEQ ID NO: 1 and the one or more second oligonucleotide probes comprise at least 20 contiguous nucleotides of SEQ ID NO: 4.

11. The kit of claim 1 , further comprising a predetermined amount of a purified ERG and/or AMACR nucleic acid.

12. The kit of claim 2 , further comprising a predetermined amount of a purified ERG and/or DD3 nucleic acid.

13. The kit of claim 8 , further comprising a predetermined amount of a purified ERG and/or DD3 nucleic acid.

14. The kit of claim 9 , further comprising a predetermined amount of a purified ERG and/or DD3 nucleic acid.

The kit of claim 8 , wherein the labeled oligonucleotide probes consist of the first labeled oligonucleotide probe comprising at least 15 contiguous nucleotides of nucleotides 510-773 of SEQ ID NO: 1 or a sequence complementary thereto, the second labeled oligonucleotide probe comprising at least 15 contiguous nucleotides of SEQ ID NO: 4 or a sequence complementary thereto, and a third labeled oligonucleotide probe that detects expression of a prostate cell-specific gene.

16. The kit of claim 1 , wherein the first and oligonucleotide probes are not affixed to a support.

17. The kit of claim 2 , wherein the wherein the first and second oligonucleotide probes are not affixed to a support.

18. The kit of claim 8 , wherein the labeled oligonucleotide probes are not affixed to a support.

19. The kit of claim 9 , wherein the one or more first and second oligonucleotide probes are not affixed to a solid support.

20. A kit for detecting prostate cancer, comprising oligonucleotide probes, wherein the oligonucleotide probes consist of a first oligonucleotide probe comprising at least 15 contiguous nucleotides of SEQ ID NO: 1 or a sequence complementary thereto, a second oligonucleotide probe comprising at least 15 contiguous nucleotides of SEQ ID NO: 4 or a sequence complementary thereto, and a third control oligonucleotide probe to compare and/or normalize expression levels of ERG and/or DD3, wherein the first, second, and third oligonucleotide probes are labeled with at least one detectable group.
