Abstract

The present invention describes a method for detecting biomolecular interactions said method comprising: (a) selecting an appropriate reporter molecule selected from the group consisting of a protein, a fluorescent protein, a luminescent protein and a phosphorescent protein; (b) effecting fragmentation of said reporter molecule such that said fragmentation results in reversible loss of reporter function; (c) fusing or attaching fragments of said reporter molecule separately to other molecules; followed by (d) reassociation of said reporter fragments through interactions of the molecules that are fused to said fragments; and (e) detecting said biomolecular interactions by reconstitution of activity of the reporter molecule.

Claims (2)

1. Molecular fragment complementation assays for the detection of molecular interactions said assays comprising a reassembly of two separate complementary fragments of a protein molecule, wherein reassembly of said two complementary fragments is operated by the interaction of molecular domains fused to each fragment of said protein molecules, and wherein detection of reassembly of said two complementary fragments is independent of other molecular processes with the proviso that said molecular fragment complementation assay is not a two hybrid assay.

2. A composition comprising two complementary fragments of a first protein molecule, wherein said two complementary fragments exhibit a detectable activity when associated, wherein each of said complementary fragments is fused to separate protein molecules with the proviso that said two complementary fragments are not ubiquitin fragments and wherein said fused products are not two hybrid constructs.
