Abstract

The present invention is directed to macrocyclic tertiary amine compounds represented by general formula (I) which are inhibitors of the beta-secretase enzyme and that are useful in the treatment of diseases in which the beta-secretase enzyme is involved, such as Alzheimer's disease. The invention is also directed to pharmaceutical compositions comprising these compounds and the use of these compounds and compositions in the treatment of such diseases in which the beta-secretase enzyme is involved.

Claims (232)

1. A compound of formula (I):

and pharmaceutically acceptable salts thereof, and individual enantiomers and diastereomers thereof, wherein:

X and Y are selected from the group consisting of

(1) hydrogen,

(2) —C 1-3 alkyl,

(3) halogen, and

(4) cyano;

A is selected from the group consisting of

(1) hydrogen,

(2) —C 1-10 alkyl,

(3) —C 2-10 alkenyl, and

(4) —C 2-10 alkynyl,

wherein said alkyl, alkenyl or alkynyl is unsubstituted or substituted with one or more

(a) halo,

(b) —C 3-8 cycloalkyl,

(c) —OH,

(d) —CN,

(e) —O—C 1-10 alkyl,

(f) —C 6-10 aryl, or

(g) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

and said aryl and heteroaryl groups are unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-10 alkyl,

(v) —C 1-10 alkyl,

(vi) —C 2-10 alkenyl,

(vii) —C 2-10 alkynyl, or

(viii) —C 3-8 cycloalkyl;

R 1 is phenylene, wherein said phenylene is unsubstituted or substituted with one or more

(a) halo,

(b) —C 1-10 alkyl,

(c) —C 2-10 alkenyl,

(d) —C 2-10 alkynyl,

(e) —OH,

(f) —CN,

(g) —O—C 1-10 alkyl, or

(h) —C 3-8 cycloalkyl;

R 2 is selected from the group consisting of:

(1) (R 5 —SO 2 )N(R 6 )—, wherein R 5 is

(a) —C 1-10 alkyl,

(b) —C 2-10 alkenyl,

(c) —C 2-10 alkynyl,

(d) —C 3-8 cycloalkyl,

(e) —C 6-10 aryl, or

(f) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

(g) —NR 7 R 8 ,

wherein said alkyl, alkenyl, alkynyl, cycloalkyl, aryl and heteroaryl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-10 alkyl,

(v) —C 1-10 alkyl,

(vi) —C 2-10 alkenyl,

(vii) —C 2-10 alkynyl,

(viii) —C 3-8 cycloalkyl,

(ix) —C 6-10 aryl, or

(x) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

and said aryl and heteroaryl is unsubstituted or substituted with one or more

(A) halo,

(B) —OH,

(C) —CN,

(D) —O—C 1-10 alkyl,

(E) —C 3-8 cycloalkyl,

(F) —C 1-10 alkyl,

(G) —C 2-10 alkenyl, or

(H) —C 2-10 alkynyl;

R 6 is selected from the group consisting of

(a) hydrogen,

(b) —C 1-10 alkyl,

(c) —C 2-10 alkenyl,

(d) —C 2-10 alkynyl,

(e) —C 6-10 aryl, or

(f) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

wherein said alkyl, alkenyl, alkynyl, aryl or heteroaryl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-10 alkyl,

(v) —C 3-8 cycloalkyl,

(vi) —C 6-10 aryl, or

(vii) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl;

wherein said cycloalkyl, aryl or heteroaryl is unsubstituted or substituted with one or more

(A) halo,

(B) —OH,

(C) —CN,

(D) —O—C 1-10 alkyl,

(E) —C 3-8 cycloalkyl, or

(F) —C 6-10 aryl,

or R 5 and R 6 may be linked to form a group —CH 2 (CH 2 ) p CH 2 —;

(2) —C 6-10 aryl, wherein said aryl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-10 alkyl,

(v) —C 3-8 cycloalkyl,

(vi) —C 1-10 alkyl,

(vi) —C 6-10 aryl, or

(4) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl,

pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl,

thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

wherein said heteroaryl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-10 alkyl,

(v) —C 3-8 cycloalkyl,

(vi) —C 1-10 alkyl,

(vii) —C(═O)—O—C 1-10 alkyl,

(viii) —C(═O)—OH, and

(ix)=—C(═O)—NR c R d ,

(x) —NR c R d , wherein R c and R d are selected from the group consisting of

 (A) hydrogen, and

 (B) —C 1-10 alkyl;

(5) hydrogen;

(6) —CF 3 ; and

(7) —O—SO 2 —R 9 ;

R 3 is

wherein R x is selected from the group consisting of

(a) hydrogen,

(b) —C 1-6 alkyl,

(c) —C 0-3 alkylene-C 3-8 cycloalkyl,

(d) —C 0-3 alkylene-C 6-10 aryl

and said R x alkyl, alkylene, cycloalkyl and aryl groups are unsubstituted or substituted with one or more

(i) halo,

(ii) —C 1-10 alkyl,

(iii) —OH,

(iv) —CN, or

 and if the dotted line leading to R y is absent, then R y is selected from the group consisting of

(a) hydrogen,

(b) —C 1-10 alkyl,

(c) —C 2-10 alkenyl,

(d) —C 2-10 alkynyl,

(e) —C 3-8 cycloalkyl,

(f) —C 0-6 alkylene-C 6-10 aryl, or

(g) —C 0-6 alkylene-heteroaryl, wherein said heteroaryl is selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

and said R y alkyl, alkylene, alkenyl, alkynyl, cycloalkyl and heteroaryl groups are unsubstituted with one or more

(i) halo,

(ii) —C 1-10 alkyl,

(iii) —OH,

(iv) —CN, or

(v) —O—C 1-10 alkyl,

 and R y′ is selected from the group consisting of

(a) hydrogen, and

(b) —CH 3 ,

 and if the dotted line leading to R y represents a bond, then R y′ is absent and R y is selected from the group consisting of

(a) ═CH—C 1-10 alkyl,

(b) ═CH—C 0-6 alkylene-C 6-10 aryl, or

(c) ═CH 2

wherein said alkyl, alkylene, cycloalkyl, aryl or heteroaryl R y groups are unsubstituted or substituted with one or more

(i) halo,

(ii) —C 1-10 alkyl,

(iii) —OH,

(iv) —CN, or

(v) —O—C 1-10 alkyl, or

(vi) —C 3-8 cycloalkyl;

 Q 3 is —CH 2 —;

R 4 is —C(═O)—O—CH 2 ;

R 7 and R 8 are selected from the group consisting of

(1) —C 1-10 alkyl, and

(2) —C 0-3 alkyene-C 6-10 aryl,

wherein said alkyl, alkylene and aryl is unsubstituted or substituted with one or more

(a) halo,

(b) —C 1-10 alkyl,

(c) —OH,

(d) —CN,

(e) —O—C 1-10 alkyl, or

(f) —C 3-8 cycloalkyl;

R 9 is selected from the group consisting of

(1) —C 1-10 alkyl, and

(2) —C 0-3 alkylene-C 6-10 aryl,

wherein said alkyl, alkylene and aryl is unsubstituted or substituted with one or more

(a) halo,

(b) —C 1-10 alkyl,

(c) —OH,

(d) —CN,

(e) —O—C 1-10 alkyl, or

(f) —C 3-8 cycloalkyl, or

R 9 is NR 7 R 8 ;

m is 1;

n is 0;

p is 1, 2, 3, 4 or 5;

q is 2, 3, 4 or 5; and

r is 0.

2. A compound of claim 1 wherein X and Y are both hydrogen.

3. A compound of claim 1 wherein the dotted line leading to R y is absent and R y is selected from the group consisting of

(a) —C 1-10 alkyl,

(b) —C 2-10 alkenyl,

(c) —C 2-10 alkynyl,

(d) —C 3-8 cycloalkyl,

(e) —C 0-6 alkylene-C 6-10 aryl, or

(f) —C 0-6 alkylene-heteroaryl, wherein said heteroaryl is selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl,

wherein said alkyl, alkylene, alkenyl, alkynyl, cycloalkyl, aryl or heteroaryl are unsubstituted or substituted with one or more

(i) halo,

(ii) —C 1-10 alkyl,

(iii) —OH,

(iv) —CN,

(v) —O—C 1-10 alkyl, or

(vi) —C 3-8 cycloalkyl,

and R y′ is hydrogen.

4. A compound of claim 1 wherein R x is hydrogen.

5. A compound of claim 1 wherein A is selected from the group consisting of

(1) hydrogen, and

(2) —C 1-10 alkyl,

wherein said alkyl is unsubstituted or substituted with one or more

(a) halo,

(b) —C 3-8 cycloalkyl,

(c) —CN

(e) —C 6-10 aryl, or

(f) heteroaryl selected from the group consisting of pyrazinyl, pyrazolyl, pyridazinyl, pyridyl, pyrimidinyl, pyrrolyl, tetrazolyl, furanyl, imidazolyl, triazinyl, pyranyl, thiazolyl, thienyl, thiophenyl, triazolyl, oxazolyl, isoxazolyl, thiazolyl, oxadiazolyl, indolyl, quinolinyl, isoquinolinyl, benzimidazolyl and benzoxazolyl.

6. A compound of claim 1 wherein R 2 is (R 5 —SO 2 )N(R 6 )—, wherein R 5 is —C 1-6 alkyl, wherein said alkyl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-6 alkyl, or

(v) —C 1-6 alkyl, and

R 6 is selected from the group consisting of

(a) hydrogen,

(b) —C 1-6 alkyl,

(c) —C 6-10 aryl,

wherein said alkyl and aryl is unsubstituted or substituted with one or more

(i) halo,

(ii) —OH,

(iii) —CN,

(iv) —O—C 1-6 alkyl or

(v) —C 1-6 alkyl.

7. A compound of claim 1 wherein R 2 is phenyl, unsubstituted or substituted with cyano.

8. A compound of claim 1 which is a compound of formula (II):

and pharmaceutically acceptable salts thereof, and individual enantiomers and diastereomers thereof.

9. A compound of claim 1 which is a compound of formula (III):

and pharmaceutically acceptable salts thereof, and individual enantiomers and diastereomers thereof.

10. A pharmaceutical composition comprising a therapeutically effective amount of a compound of claim 1 and a pharmaceutically acceptable carrier.
