Abstract

The present invention relates to methods and compositions for predicting drug responses. In particular, the present invention provides methods and compositions for determining individualized Warfarin dosages based on genotype of DNA polymorphisms and haplotypes derived from them in the VKORC1 gene.

Claims (36)

1. A method of predicting a human subject's response to warfarin, said method comprising:

providing a sample from a human subject;

determining, in the subject's VKORC1 gene, the presence of an adenosine or guanine at the nucleotide corresponding to position 3673 of SEQ ID NO:1 in the sample from the subject; and

predicting, based on the subject's genotype of A/A, A/G, or G/G at position 3673, the human subject's response to warfarin.

2. The method according to claim 1 , wherein said determining comprises:

sequencing a nucleic acid molecule from the sample at least the nucleotide corresponding to position 3673 of SEQ ID NO: 1.

3. The method according to claim 1 , wherein said determining comprises:

detecting, in a hybridization assay, an ability of a nucleic acid molecule from the sample to hybridize to an oligonucleotide probe.

4. The method according to claim 1 , wherein said determining comprises:

detecting, in a PCR-based assay, an ability of oligonucleotide primers to amplify a nucleic acid molecule from the sample.

5. The method according to claim 1 , wherein the nucleotide corresponding to position 3673 is adenosine.

6. The method according to claim 1 , wherein the nucleotide corresponding to position 3673 is guanine.

7. A method of determining a suitable dose of warfarin for a human subject, said method comprising:

providing a sample from a human subject;

determining, in the subject's VKORC1 gene, the presence of an adenosine or guanine at the nucleotide corresponding to position 3673 of SEQ ID NO:1 in the sample from the subject; and

identifying a suitable dose of warfarin for the subject based on the subject's genotype of A/A, A/G, or G/G at position 3673.

8. The method according to claim 7 , wherein said determining comprises:

sequencing a nucleic acid molecule from the sample at least the nucleotide corresponding to position 3673 of SEQ ID NO: 1.

9. The method according to claim 7 , wherein said determining comprises:

detecting, in a hybridization assay, an ability of a nucleic acid molecule from the sample to hybridize to an oligonucleotide probe.

10. The method according to claim 7 , wherein said determining comprises:

detecting, in a PCR-based assay, an ability of oligonucleotide primers to amplify a nucleic acid molecule from the sample.

11. The method according to claim 7 , wherein the nucleotide corresponding to position 3673 is adenosine.

12. The method according to claim 7 , wherein the nucleotide corresponding to position 3673 is guanine.

13. The method according to claim 7 further comprising:

administering warfarin to the subject at a dosage determined from said identifying.

14. A method comprising:

providing a sample from a human subject receiving warfarin therapy;

determining, in the subject's VKORC1 gene, the presence of an adenosine or guanine at the nucleotide corresponding to position 3673 of SEQ ID NO:1 in the sample from the subject; and

establishing a correlation between the subject's genotype of A/A, A/G, or G/G at position 3673 with the subject's maintenance warfarin dose.

15. The method according to claim 14 , wherein the nucleotide corresponding to position 3673 is adenosine.

16. The method according to claim 14 , wherein the nucleotide corresponding to position 3673 is guanine.

17. A method of predicting a dose range of warfarin for a human patient, comprising:

investigating the nucleotide at the 3673 position of the VKORC1 gene of the patient; wherein the nucleotide is investigated by using an oligonucleotide that specifically hybridizes with the promoter of the VKORC1 gene;

determining whether the patient requires a higher or lower dose of warfarin based on the nucleotide at the 3673 position, wherein homozygous AA, heterozygous AG, and homozygous GG at this position indicate that the patient requires a lower, intermediate, or higher warfarin dose respectively; and

predicting a warfarin dosage range based on said determining whether the patient requires a higher or lower dose of warfarin.
