Abstract

The present invention is in the field of plant breeding and genetics, particularly as it pertains to the genus, Glycine . More specifically, the invention relates to a method for screening soybean plants containing one or more quantitative trait loci for disease resistance, species of Glycine having such loci and methods for breeding for and screening of Glycine with such loci. The invention further relates to the use of exotic germplasm in a breeding program.

Claims (24)

1. A method for assaying a soybean plant leaf for disease resistance, immunity, or susceptibility comprising the steps of:

a. obtaining a part of said soybean plant leaf;

b. cultivating said part in a media consisting essentially of water, wherein said media is capable of maintaining said part for an assay for up to 2 months;

c. exposing said part to a fungal or bacterial plant leaf pathogen; and

d. assessing said part for resistance, immunity, or susceptibility to disease caused by said fungal or bacterial plant leaf pathogen.

2. The method of claim 1 , wherein said leaf is selected from the group consisting of a unifoliate leaf, a V1 trifoliate leaf, a V2 trifoliate leaf and a V3 trifoliate leaf.

The method of claim 1 , wherein said exposing said part to a fungal or bacterial plant pathogen is accomplished by a method selected from the group consisting of (a) direct application of the pathogen to the tissue; (b) inclusion of the pathogen in the culture media; and (c) inclusion of an agent that is effectively contaminated with the pathogen and serves to inoculate the tissue.

4. The method of claim 1 , wherein said fungal or bacterial plant pathogen exposure can be in the form of pathogen macromolecules, cells, tissues, whole organism or combinations thereof, and the pathogen, or parts thereof, mediates an immune response in said part.

The method of claim 1 , wherein said disease resistance, immunity, or susceptibility comprises a reaction to a fungal or bacterial pathogen selected from the group consisting of Phakopsora pachyrhizi, Phakopsora meibomiae (Asian Soybean Rust), Colletotrichum dematium var. truncatum, Sclerotinia sclerotiorum (Sclerotinia stem rot), Septoria glycines , (Brown Spot), Phomopsis Ion gicola (Stem blight), Phomopsis spp. (Phomopsis seed decay), Peronospora manshurica (Downy Mildew), Rhizoctonia solani (Rhizoctonia root and stem rot, Rhizoctonia aerial blight), Cercospora kikuchii (Purple Seed Stain), Alternaria sp. (Target Spot), Cercospora sojina (Frogeye Leafspot), Corynespora cassiicola (Target spot), Pseudomonas savastonoi pv. glycinea (Bacterial blight), Xanthomonas axonopodis pv. glycines , (Bacterial pustule), and Curtobacterium flaccumfaciens pv. flaccumfaciens , (Bacterial tan spot).

The method of claim 1 , wherein said part is assessed for resistance, immunity, or susceptibility to Phakopsora pachyrhizi, Phakopsora meibomiae (Asian Soybean Rust), Colletotrichum dematium var. truncatum, Sclerotinia sclerotiorum (Sclerotinia stem rot), Septoria glycines , (Brown Spot), Phomopsis longicola (Stem blight), Phomopsis spp. (Phomopsis seed decay), Peronospora manshurica (Downy Mildew), Rhizoctonia solani (Rhizoctonia root and stem rot, Rhizoctonia aerial blight), Corynespora cassiicola (Target spot), Pseudomonas savastonoi pv. glycinea (Bacterial blight), Xanthomonas axonopodis pv. glycines , (Bacterial pustule), and Curtobacterium flaccumfaciens pv. flaccumfaciens , (Bacterial tan spot).

The method of claim 1 , wherein said selected soybean plant is selected from the group consisting of Glycine arenaria, Glycine argyrea, Glycine canescens, Glycine clandestine, Glycine curvata, Glycine cyrtoloba, Glycine falcate, Glycine latifolia, Glycine latrobeana, Glycine max, Glycine microphylla, Glycine pescadrensis, Glycine pindanica, Glycine rubiginosa, Glycine soja, Glycine sp., Glycine stenophita, Glycine tabacina and Glycine tomentella.

8. The method of claim 1 , wherein said media consists of water.

9. The method of claim 1 , wherein said disease resistance, immunity, or susceptibility is selected from the group consisting of Cercospora sojina (Frogeye leaf spot), Sclerotinia (stemrot), and Asian Soybean Rust (ASR) resistance.

10. The method of claim 1 , wherein said soybean plant is a progeny from a cross between a soybean plant comprising a desirable disease resistance quantitative trait locus and a soybean plant of an elite line.

11. A soybean plant selected using the method of claim 1 .

12. A method for assaying a soybean plant leaf for disease resistance, immunity, or susceptibility to a fungal or bacterial leaf disease comprising the steps of:

a. obtaining a part of said soybean plant leaf;

b. cultivating said part in a media consisting essentially of water and nutrients necessary to sustain infection, wherein said media is capable of maintaining said part for an assay for up to 2 months;

c. exposing said part to a plant leaf pathogen; and

d. assessing said part for resistance, immunity, or susceptibility to disease caused by said plant leaf pathogen.

13. The method of claim 12 , wherein said part is a leaf section.

14. The method of claim 12 , wherein said part is cultured on filter paper with the stomatal side up.

15. The method of claim 12 , wherein said media consists of water and nutrients.

16. The method of claim 12 , wherein said assay is high throughput.
