Abstract

The present invention provides a plant transformation vector for producing a plant for detecting an environmental chemical by a change of flower color, the plant transformation vector comprising an AhR gene expressibly incorporated therein, an AhR ligand stimulus-inducible promoter, and an expression suppressive factor for suppressing the expression of a gene involved in the synthesis of a desired flower pigment, the factor being expressibly incorporated therein by the promoter.

Claims (16)

1 . A plant transformation vector for producing a plant for detecting an environmental chemical by a change of flower color, the plant transformation vector comprising an AhR gene expressibly incorporated therein, an AhR ligand stimulus-inducible promoter, and an expression suppressive factor for suppressing expression of a gene involved in the synthesis of a desired flower pigment, the factor being expressibly incorporated therein by the promoter.

The plant transformation vector according to claim 1 , comprising genes coding for a DNA-binding domain, an AhR ligand-binding domain and a transcriptional activation domain, also comprising a nucleic acid coding for a double strand RNA having an RNA interference effect on the gene involved in the synthesis of the flower pigment; wherein the nucleic acid is under the control of the AhR ligand stimulus-inducible promoter, and the DNA-binding domain binds the AhR ligand stimulus-inducible promoter in response to an AhR ligand.

3 . The plant transformation vector according to claim 1 , wherein the gene involved in the synthesis of the flower pigment is a gene involved in the biosynthesis of anthocyanin.

4 . The plant transformation vector according to claim 3 , wherein the gene involved in the biosynthesis of anthocyanin is a gene coding for flavanone 3-hydroxylase (F3H) or flavonoid 3′,5′-hydroxylase (F3′,5′H).

5 . The plant transformation vector according to claim 1 , wherein an AhR ligand is any of dioxins/dioxin-like compounds, and/or polycyclic aromatic hydrocarbons, or an analog thereof.

6 . The plant transformation vector according to claim 1 , comprising XD 4 V or XD 5 V.

7 . The plant transformation vector according to claim 1 , wherein the AhR ligand stimulus-inducible promoter includes a LexA promoter sequence.

8 . The plant transformation vector according to claim 1 , further comprising at least one desired gene.

9 . The plant transformation vector according to claim 8 , wherein the desired gene is a drug resistance gene.

10 . The plant transformation vector according to claim 8 , wherein the desired gene is carnation THC2′GT gene.

11 . The plant transformation vector according to claim 1 , wherein the plant is petunia, torenia or verbena.

12 . A transgenic plant for changing the flower color thereof in response to an AhR ligand, the transgenic plant being transformed with the vector according to claim 1 .

13 . The transgenic plant according to claim 12 , which is petunia, torenia or verbena.

14 . A reproduction material of the transgenic plant according to claim 12 .

15 . A method for detecting an AhR ligand in soil using a transgenic plant, the method comprising the step of transplanting the transgenic plant according to claim 12 to soil to be tested, cultivating the transgenic plant, and observing a change of flower color.

16 . The method according to claim 15 , wherein the AhR ligand is any of dioxins/dioxin-like compounds, and/or polycyclic aromatic hydrocarbons, or an analog thereof.
