Abstract

The present invention relates to a novel class of modified malonate derivatives. The modified malonate compounds can be used to treat cancer. The modified malonate compounds can also inhibit histone deacetylase and are suitable for use in selectively inducing terminal differentiation, and arresting cell growth and/or apoptosis of neoplastic cells, thereby inhibiting proliferation of such cells. Thus, the compounds of the present invention are useful in treating a patient having a tumor characterized by proliferation of neoplastic cells. The compounds of the invention may also be useful in the prevention and treatment of TRX-mediated diseases, such as autoimmune, allergic and inflammatory diseases, and in the prevention and/or treatment of diseases of the central nervous system (CNS), such as neurodegenerative diseases. The present invention further provides pharmaceutical compositions comprising the modified malonate derivatives and safe dosing regimens of these pharmaceutical compositions, which are easy to follow, and which result in a therapeutically effective amount of the modified malonate derivatives in vivo.

Claims (266)

1. A compound represented by the following structural Formula:

wherein Y 1 and Y 2 are, independently of each other, hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

w is 1 or 2, wherein when w is 2, the two Y 1 are the same or different;

z is 1 or 2, wherein when z is 2, the two Y 2 are the same or different;

v, j, k, g are independently of each other 0, 1, 2, 3 or 4, wherein v+j+k+g is less or equal to 4;

h, q, m, r are independently of each other 0, 1, 2, 3 or 4, wherein h+q+m+r is less or equal to 4;

when j is not 0, A 2 is selected from —NH—, —S—, —O—, ethenyl or a bond; when j is 0, A 2 is a bond or ethenyl;

when m is not 0, A 1 is selected from —NH—, —S—, —O—, ethenyl or a bond; when m is 0, A 1 is a bond or ethenyl;

n is 0, 1, 2, 3 or 4;

R 1 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl, halo, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

R 2 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl or halo;

s is 1 or 2, and when s is 2, R 2 is the same or different;

R 9 , R 10 , R 13 and R 14 are independently of each other selected from hydrogen or C 1 -C 4 alkyl;

R 11 and R 12 are independently of each other selected from hydrogen, C 1 -C 4 alkyl, or halo;

R 8 and R 15 are independently of each other selected from hydrogen, C 1 -C 4 alkyl, OH, or halo;

R 6 and R 7 are independently of each other hydrogen or C 1 -C 4 alkyl;

Ring B is cycloalkyl, heteroaryl, heterocyclic or aryl;

R 3 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, —NHSO 2 , —SO 2 NH, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—, C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino;

t is 1, 2, 3 or 4;

L 1 is (CH 2 ) r , ethenyl or cyclopropyl, wherein r is 0, 1 or 2;

Ring A is a 5 to 6 membered heteroaryl or aryl;

X is OH, SH or NH 2 ;

p is 1,2, 3 or 4;

R 5 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—, C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino or L 2 -R 16 , wherein R 16 is substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic, substituted or unsubstituted aryl, substituted or unsubstituted C 3 -C 8 cycloalkyl, L 2 is selected from a bond, C 1 -C 4 alkylene, C 1 -C 4 alkynyl, C 1 -C 4 alkenyl, —O—, —S—, —N—, —C(═O)NH—, —NHC(═O)—, —NHC(═O)NH—, —SO 2 NH—, —NHSO 2 —, —SO 2 —, —C(═O)— or —C(═O)O—;

or a stereoisomer or pharmaceutically acceptable salt thereof.

2. The compound of claim 1 represented by the following structural Formula:

wherein Y 1 and Y 2 are, independently of each other, hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

w is 1 or 2, wherein when w is 2, the two Y 1 are the same or different;

z is 1 or 2, wherein when z is 2, the two Y 2 are the same or different;

j and m are independently of each other 0, 1, 2, 3 or 4;

when j is not 0, A 2 is selected from —NH—, —S—, —O—, ethenyl or a bond; when j is 0, A 2 is a bond or ethenyl;

when m is not 0, A 1 is selected from —NH—, —S—, —O—, ethenyl or a bond; when m is 0, A 1 is a bond or ethenyl;

n is 0, 1, 2, 3 or 4;

R 1 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl, halo, substituted or unsubstituted cycloalkyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

R 2 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl or halo;

s is 1 or 2, and when s is 2, R 2 is the same or different;

R 9 and R 14 are independently of each other selected from hydrogen or C 1 -C 4 alkyl;

Ring B is cycloalkyl, heteroaryl, heterocyclic or aryl;

R 3 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, —NHSO 2 , —SO 2 NH, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino;

t is 1,2, 3 or 4;

L 1 is (CH 2 ) r , cyclopropyl, or ethenyl, wherein r is 0, 1 or 2;

Ring A is a 5 to 6 membered heteroaryl or aryl;

X is OH, SH or NH 2 ;

p is 1,2, 3 or 4;

R 5 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—, C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino or L 2 -R 16 , wherein R 16 is substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic, substituted or unsubstituted aryl, substituted or unsubstituted C 3 -C 8 cycloalkyl, L 2 is selected from a bond, C 1 -C 4 alkylene, C 1 -C 4 alkynyl, C 1 -C 4 alkenyl, —O—, —S—, —N—, —C(═O)NH—, —NHC(═O)—,

3. The compound of claim 1 represented by the following structural formula:

wherein Y 1 and Y 2 are, independently of each other, hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

w is 1 or 2, wherein when w is 2, the two Y 1 are the same or different;

z is 1 or 2, wherein when z is 2, the two Y 2 are the same or different;

j and m are independently of each other 0, 1, 2, 3 or 4;

when j is not 0, A 2 is selected from —NH—, —S—, —O—, ethenyl or a bond; when k is 0, A 2 is a bond or ethenyl;

when m is not 0, A 1 is selected from —NH—, —S—, —O—, ethenyl or a bond; when m is 0, A 1 is a bond or ethenyl;

n is 0, 1, 2, 3 or 4;

R 1 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl, halo, substituted or unsubstituted aryl;

R 2 is selected from hydrogen, OH, NH 2 , C 1 -C 4 alkyl or halo;

R 9 and R 14 are independently of each other selected from hydrogen or C 1 -C 4 alkyl;

Ring B is heteroaryl or aryl;

R 3 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—, C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino;

R 17 , R 18 and R 20 are independently of each other selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, —NHSO 2 , —SO 2 NH, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—, C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino;

R 19 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 7 alkoxy, C 1 -C 7 alkyl, C 1 -C 7 haloalkyl, C 1 -C 7 haloalkyloxy, C 1 -C 7 hydroxyalkyl, C 1 -C 7 alkenyl, C 1 -C 7 alkyl-C(═O)O—, C 1 -C 7 alkyl-C(═O)—, C 1 -C 7 alkynyl, halo group, amide, hydroxyalkoxy, —NHSO 2 , —SO 2 NH, C 1 -C 7 alkyl-NHSO 2 —, C 1 -C 7 alkyl-SO 2 NH—C 1 -C 7 alkylsulfonyl, C 1 -C 7 alkylamino or di(C 1 -C 7 )alkylamino or L 2 -R 16 , wherein R 16 is substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic, substituted or unsubstituted aryl, substituted or unsubstituted C 3 -C 8 cycloalkyl, L 2 is selected from a bond, C 1 -C 4 alkylene, C 1 -C 4 alkynyl, C 1 -C 4 alkenyl, —O—, —S—, —N—, —C(═O)NH—, —NHC(═O)—,

or a stereoisomer or pharmaceutically acceptable salt thereof.

4. The compound of claim 3 , wherein Y 1 and Y 2 are, independently of each other, a substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted heteroaryl, or substituted or unsubstituted aryl.

The compound of claim 4 , wherein Y 1 and Y 2 are, independently of each other, an optionally substituted phenyl, indolyl, quinolinyl, pyridyl, biphenyl, naphthyl, thiazolyl, pyrrolidinyl, benzodioxinyl, said optional substituent comprises one, two or three groups selected from hydroxyl, halo, amino, nitro, CN, C 1 -C 4 alkyl, C 1 -C 4 alkoxy, C 1 -C 4 haloalkyl, C 1 -C 4 alkylamino, di(C 1 -C 4 )alkylamino, C 1 -C 4 alkyl-NHS(O) 2 —, C 1 -C 4 alkyl-S(O) 2 NH—, aryl C 1 -C 4 alkoxy, heterocyclic, aryl or heteroaryl.

6. The compound of claim 3 , wherein Ring B is selected from phenyl, benzothiophenyl, benzofuranyl, thiazolyl, benzothiazolyl, furanyl, pyridyl, pyrimidyl, quinolinyl, thiophenyl, benzodioxyl, benzooxadiazolyl, quinoxalinyl, benzotriazolyl, benzoimidazolyl or benzooxazolyl.

The compound of claim 3 , wherein R 19 is selected from hydrogen, OH, NH 2 , nitro, CN, amide, carboxyl, C 1 -C 4 alkoxy, C 1 -C 4 alkyl, C 1 -C 4 haloalkyl, C 1 -C 4 hydroxyalkyl, C 1 -C 4 alkenyl, C 1 -C 4 alkyl-C(═O)O—, C 1 -C 4 alkynyl, halo group, amide or L 2 -R 16 , wherein R 16 is substituted or unsubstituted heteroaryl, substituted or unsubstituted aryl, L 2 is selected from a bond, ethylene, ethenyl, ethynyl, —O—, —S—, —C(═O)NH—, —NHC(═O)—, —C(═O)— or —C(═O)O—.

8. The compound of claim 7 , wherein R 17 , R 18 and R 20 are independently of each other hydrogen or fluoro.

9. The compound of claim 8 , wherein R 19 is hydrogen or phenyl and X is amino.

10. The compound of claim 3 , wherein w and z are 1, R 9 and R 14 are hydrogen, and A 1 and A 2 are each a bond.

11. The compound of claim 1 represented by the following structural formula:

wherein Y 1 and Y 2 are, independently of each other, hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

w is 1 or 2, wherein when w is 2, the two Y 1 are the same or different;

z is 1 or 2, wherein when z is 2, the two Y 2 are the same or different;

j and m are independently of each other 0, 1, 2, 3 or 4;

when j is not 0, A 2 is selected from —NH—, —S—, —O—, ethenyl or a bond; when k is 0, A 2 is a bond or ethenyl;

when m is not 0, A 1 is selected from —NH—, —S—, —O—, ethenyl or a bond; when m is 0, A 1 is a bond or ethenyl;

12. A compound represented by the following structural Formula:

wherein Y 1 is selected from hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl; or Y 1 and R 7 are combined to form a substituted or unsubstituted C 1 -C 10 alkylene;

wherein Y 2 is selected from hydrogen, a substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 1 -C 10 alkyl, substituted or unsubstituted C 1 -C 10 alkenyl, substituted or unsubstituted heteroaryl, substituted or unsubstituted heterocyclic or substituted or unsubstituted aryl;

or Y 2 and R 6 are combined to form a substituted or unsubstituted C 1 -C 10 alkylene;

w is 1 or 2, wherein when w is 2, the two Y 1 are the same or different;

z is 1 or 2, wherein when z is 2, the two Y 2 are the same or different;

j and m are independently of each other 0, 1, 2, 3 or 4;

when j is not 0, A 2 is selected from —NH—, —S—, —O—, ethenyl or a bond; when j is 0, A 2 is a bond or ethenyl;

when m is not 0, A 1 is selected from —NH—, —S—, —O—, ethenyl or a bond; when m is 0, A 1 is a bond or ethenyl;

n is 0, 1, 2, 3 or 4;
