Abstract

Compounds, pharmaceutical compositions, kits and methods are provided for use with MEK that comprise a compound selected from the group consisting of: wherein the variables are as defined herein.

Claims (122)

1. A compound represented by the formula:

or an ester, tautomer, enantiomer, pharmaceutically acceptable salt or prodrug thereof,

wherein

R 1 is selected from the group consisting of (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl, (C 9-12 )bicycloalkyl, hetero(C 3-12 )bicycloalkyl, (C 4-12 )aryl, hetero(C 1-10 )aryl, (C 9-12 )bicycloaryl and hetero(C 4-12 )bicycloaryl, each substituted or unsubstituted;

R 3 is selected from the group consisting of hydroxy, amino, (C 1-10 )alkylamino, (C 1-10 )alkoxyamino, hydroxy(C 1-10 )alkoxyamino, hydroxy(C 1-10 )alkylamino, cycloamino, sulfonamido, imino, (C 2-10 )alkyl, halo(C 1-10 )alkyl, carbonyl(C 1-3 )alkyl, thiocarbonyl(C 1-3 )alkyl, sulfonyl(C 1-3 )alkyl, sulfinyl(C 1-3 )alkyl, amino (C 1-10 )alkyl, imino(C 1-3 )alkyl, (C 3-12 )cycloalkyl(C 1-5 )alkyl, hetero(C 3-12 )cycloalkyl(C 1-5 )alkyl, aryl(C 1-10 )alkyl, heteroaryl(C 1-5 )alkyl, (C 9-12 )bicycloaryl(C 1-5 )alkyl, hetero(C 8-12 )bicycloaryl(C 1-5 )alkyl, (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl, (C 9-12 )bicycloalkyl, hetero(C 3-12 )bicycloalkyl, (C 4-12 )aryl, hetero(C 1-10 )aryl, (C 9-12 )bicycloaryl and hetero(C 4-12 )bicycloaryl, each substituted or unsubstituted;

R 4 is selected from the group consisting of amino, (C 1-5 )alkylamino, (C 1-5 )alkyl, halo(C 1-5 )alkyl, hydroxy(C 1-5 )alkyl, (C 1-8 )azaalkyl, hydroxy(C 1-8 )azaalkyl and (C 3-6 )cycloalkyl, each substituted or unsubstituted;

R 5 is selected from the group consisting of hydrogen, (C 1-5 )alkyl, amino (C 1-10 )alkyl, hydroxy(C 1-10 )alkyl and (C 3-12 )cycloalkyl, each substituted or unsubstituted; and

R 7 is selected from the group consisting of hydrogen, halo, nitro, cyano, thio, oxy, hydroxy, carbonyloxy, (C 1-10 )alkoxy, (C 4-12 )aryloxy, hetero(C 1-10 )aryloxy, carbonyl, oxycarbonyl, aminocarbonyl, amino, (C 1-10 )alkylamino, sulfonamido, imino, sulfonyl, sulfinyl, (C 1-10 )alkyl, halo(C 1-10 )alkyl, hydroxy(C 1-10 )alkyl, carbonyl(C 1-10 )alkyl, thiocarbonyl(C 1-10 )alkyl, sulfonyl(C 1-10 )alkyl, sulfinyl(C 1-10 )alkyl, (C 1-10 )azaalkyl, imino(C 1-10 )alkyl, (C 3-12 )cycloalkyl(C 1-5 )alkyl, hetero(C 3-12 )cycloalkyl(C 1-10 )alkyl, aryl(C 1-10 )alkyl, hetero(C 1-10 )aryl(C 1-5 )alkyl, (C 9-12 )bicycloaryl(C 1-5 )alkyl, hetero(C 8-12 )bicycloaryl(C 1-5 )alkyl, (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl, (C 9-12 )bicycloalkyl, hetero(C 3-12 )bicycloalkyl, (C 4-12 )aryl, hetero(C 1-10 )aryl, (C 9-12 )bicycloaryl and hetero(C 4-12 )bicycloaryl, each substituted or unsubstituted.

2. A compound of claim 1 represented by the formula:

or an ester, tautomer, enantiomer, pharmaceutically acceptable salt or prodrug thereof,

wherein

R 11 is hydrogen, a substituent convertible in vivo to hydrogen, a substituted or unsubstituted (C 1-10 )alkyl, or a substituted or unsubstituted halo(C 1-10 )alkyl; and

R 12 is selected from the group consisting of hydrogen, oxy, hydroxy, alkoxy, aryloxy, heteroaryloxy, carbonyl, amino, (C 1-10 )alkylamino, sulfonamido, imino, sulfonyl, sulfinyl, (C 1-10 )alkyl, halo(C 1-10 )alkyl, carbonyl(C 1-3 )alkyl, thiocarbonyl(C 1-3 )alkyl, sulfonyl(C 1-3 )alkyl, sulfinyl(C 1-3 )alkyl, amino (C 1-10 )alkyl, imino(C 1-3 )alkyl, (C 3-12 )cycloalkyl(C 1-5 )alkyl, hetero(C 3-12 )cycloalkyl(C 1-5 )alkyl, aryl(C 1-10 )alkyl, heteroaryl(C 1-5 )alkyl, (C 9-12 )bicycloaryl(C 1-5 )alkyl, hetero(C 8-12 )bicycloaryl(C 1-5 )alkyl, (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl, (C 9-12 )bicycloalkyl, hetero (C 3-12 )bicycloalkyl, (C 4-12 )aryl, hetero(C 1-10 )aryl, (C 9-12 )bicycloaryl and hetero(C 4-12 )bicycloaryl, each substituted or unsubstituted, or R 11 and R 12 are taken together to form a substituted or unsubstituted ring.

3. A compound of claim 1 represented by the formula:

or an ester, tautomer, enantiomer, pharmaceutically acceptable salt or prodrug thereof,

wherein

R 11 is hydrogen, a substituent convertible in vivo to hydrogen, a substituted or unsubstituted (C 1-10 )alkyl, or a substituted or unsubstituted halo(C 1-10 )alkyl; and

R 13 is selected from the group consisting of hydrogen, (C 1-10 )alkyl, halo(C 1-10 )alkyl, carbonyl(C 1-3 )alkyl, thiocarbonyl(C 1-3 )alkyl, sulfonyl(C 1-3 )alkyl, sulfinyl(C 1-3 )alkyl, amino (C 1-10 )alkyl, imino(C 1-3 )alkyl, (C 3-12 )cycloalkyl(C 1-5 )alkyl, hetero(C 3-12 )cycloalkyl(C 1-5 )alkyl, aryl(C 1-10 )alkyl, heteroaryl(C 1-5 )alkyl, (C 9-12 )bicycloaryl(C 1-5 )alkyl, hetero(C 8-12 )bicycloaryl(C 1-5 )alkyl, (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl, (C 9-12 )bicycloalkyl, hetero(C 3-12 )bicycloalkyl, (C 4-12 )aryl, hetero(C 1-10 )aryl, (C 9-12 )bicycloaryl and hetero(C 4-12 )bicycloaryl, each substituted or unsubstituted, or R 13 and R 11 are taken together to form a substituted or unsubstituted ring.

4. The compound according to any of claims 1 , 2 , or 3 , wherein R 1 is selected from the group consisting of (C 4-12 )aryl and hetero(C 1-10 )aryl, each substituted or unsubstituted.

5. The compound according to any of claims 1 , 2 , or 3 , wherein R 1 is a substituted or unsubstituted (C 4-12 )aryl.

6. The compound according to any of claims 1 , 2 , or 3 , wherein R 1 is a substituted or unsubstituted phenyl.

7. The compound according to claim 5 , wherein R 5 is a substituted or unsubstituted (C 1-5 )alkyl.

8. The compound according to claim 5 , wherein R 5 is hydrogen.

9. The compound according to claim 5 , wherein R 5 is methyl.

10. The compound according to claim 5 , wherein R 7 is hydrogen.

11. The compound according to claim 5 , wherein R 7 is a substituted or unsubstituted (C 1-3 )alkyl.

12. The compound according to claim 5 , wherein R 7 is halo.

13. The compound according to claim 2 , wherein R 11 is hydrogen.

14. The compound according to claim 2 , wherein R 11 is selected from the group consisting of (C 1-5 )alkyl and halo(C 1-5 )alkyl, each substituted or unsubstituted.

15. The compound according to claim 2 , wherein R 12 is selected from the group consisting of hydrogen, hydroxyl, alkoxy, (C 1-10 )alkyl, hydroxy(C 1-10 )alkyl, (C 3-12 )cycloalkyl, hetero(C 3-12 )cycloalkyl and hetero(C 1-10 )aryl, each substituted or unsubstituted.

16. The compound according to claim 3 , wherein R 13 is selected from the group consisting of hydrogen, alkoxy, (C 1-10 )alkyl, hydroxy(C 1-10 )alkyl and hetero(C 3-12 )cycloalkyl(C 1-10 )alkyl, each substituted or unsubstituted.

17. The compound according to claim 1 selected from the group consisting of:

5-Acetyl-2-(2-fluoro-4-iodo-phenylamino)-1H-pyrrole-3-carboxylic acid;

5-Acetyl-2-(2-fluoro-4-iodophenylamino)-N-(2-hydroxyethoxy)-1H-pyrrole-3-carboxamide;

18. The compound according to claim 1 selected from the group consisting of:

5-Acetyl-2-(2-fluoro-4-iodophenylamino)-N-(2-hydroxyethoxy)-1-methyl-1H -pyrrole-3-carboxamide;

(R)-5-Acetyl-N-(2,3-dihydroxypropoxy)-2-(2-fluoro-4-iodophenylamino)-1-methyl-1H-pyrrole-3-carboxamide;

(S)-5-Acetyl-N-(2,3-dihydroxypropoxy)-2-(2-fluoro-4-iodophenylamino)-1-methyl-1H-pyrrole-3-carboxamide;

(R)-5-Acetyl-4-chloro-N-(2,3-dihydroxypropoxy)-2-(2-fluoro-4-iodophenylamino)-1-methyl-1H-pyrrole-3-carboxamide;

(S)-5-Acetyl-4-chloro-N-(2,3-dihydroxypropoxy)-2-(2-fluoro-4-iodophenylamino)-1-methyl-1H-pyrrole-3-carboxamide;

5-Acetyl-N-(1,3-dihydroxypropan-2-yloxy)-2-(2-fluoro-4-iodophenylamino)-1-methyl-1H-pyrrole-3-carboxamide;

19. The compound according to claim 1 , wherein the compound is in the form of a pharmaceutically acceptable salt.

20. The compound according to claim 1 , wherein the compound is present in a mixture of stereoisomers.

21. The compound according to claim 1 , wherein the compound is a single stereoisomer.

22. A pharmaceutical composition comprising as an active ingredient a compound according to claim 1 and a pharmaceutical excipient.

23. The pharmaceutical composition according to claim 22 , wherein the composition is a solid formulation adapted for oral administration.

24. The pharmaceutical composition according to claim 22 , wherein the composition is a liquid formulation adapted for oral administration.

25. The pharmaceutical composition according to claim 22 , wherein the composition is a tablet.

26. The pharmaceutical composition according to claim 22 , wherein the composition is a liquid formulation adapted for parenteral administration.

27. A pharmaceutical composition comprising a compound according to claim 1 , wherein the composition is adapted for administration by a route selected from the group consisting of orally, parenterally, intraperitoneally, intravenously, intraarterially, transdermally, sublingually, intramuscularly, rectally, transbuccally, intranasally, liposomally, via inhalation, vaginally, intraoccularly, via local delivery, subcutaneously, intraadiposally, intraarticularly, and intrathecally.

28. A method of inhibiting a Mitogen-Activated Protein Kinase (MEK) comprising contacting the MEK with a compound according to claim 1 .

29. A method of inhibiting Mitogen-Activated Protein Kinase (MEK) comprising causing a compound according to claim 1 to be present in a subject in order to inhibit the MEK in vivo.

30. A method of inhibiting Mitogen-Activated Protein Kinase (MEK) comprising administering a prodrug of a compound according to claim 1 to a subject.
