Abstract

RNA aptamers and methods for identifying the same are disclosed. The RNA aptamers selectively bind coagulation factors, E2F family members, Ang1 or Ang2, and therapeutic and other uses for the RNA aptamers are also disclosed.

Claims (27)

1. A method of modulating the biological activity of a coagulation pathway factor IX or IXa comprising administering an effective amount of a RNA aptamer that binds to the coagulation pathway factor IX or IXa to a host, wherein the aptamer comprises a secondary structure consisting essentially of, in a 5′ to 3′ direction, a first stem region, a first loop region, a second stem region, a second loop region, and a third loop region, wherein the first loop region comprises a consensus sequence comprising NNAUA, wherein N is selected from the group consisting of A, U, G, and C.

2. The method of claim 1 , wherein the aptamer has a dissociation constant of about 20 nanomolar (nm) or less.

3. The method of claim 2 , wherein the dissociation constant ranges from about 400 picomolar (pM) to about 10 nM.

4. The method of claim 2 , wherein the dissociation constant ranges from about 100 pM to about 10 nM.

5. The method of claim 1 , wherein the aptamer comprises at least one modified nucleotide.

6. The method of claim 5 , wherein the aptamer comprises at least one 2′-modified nucleotide.

7. The method of claim 5 , wherein the aptamer comprises at least one 2′-halo-modified nucleotide.

8. The method of claim 5 , wherein the aptamer comprises at least one 2′-fluoro-modified nucleotide.

9. The method of claim 5 , wherein the aptamer comprises at least one 2′-O-alkyl-modified nucleotide.

10. The method of claim 5 , wherein the aptamer comprises at least one 2′-methoxy-modified nucleotide.

11. The method of claim 5 , wherein at least one cytidine is 2′deoxy-2′fluorocytidine.

12. The method of claim 5 , wherein at least one uridine is 2′deoxy- 2′-fluorouridine.

13. The method of claim 1 , wherein the aptamer comprises a 3′ chain terminator.

14. The method of claim 1 , wherein the aptamer comprises about 15 to 100 bases.

15. The method of claim 1 , wherein the aptamer comprises less than 100 bases.

16. The method of claim 15 , wherein the aptamer comprises less than 40 bases.

17. The method of claim 1 , wherein the aptamer comprises a covalently linked carrier.

18. The method of claim 17 , wherein the carrier is a soluble polymer.

19. The method of claim 17 , wherein the carrier is a biodegradable polymer.

20. The method of claim 17 , wherein the carrier is polyethylene glycol.

21. The method of claim 1 , wherein the aptamer additionally comprises a covalently linked cholesterol.

22. The method of claim 1 , wherein the first stem region comprise at least about 5 nucleotides at a 5′ end of the aptamer that form base pairs with at least about 5 nucleotides at a 3′ end of the aptamer.

23. The method of claim 1 , wherein the aptamer comprises SEQ ID NO: 3 or a truncate thereof.

24. The method of claim 1 , wherein the aptamer comprises a nucleotide sequence at least 80% homologous to SEQ ID NO: 3.

25. The method of claim 1 , wherein the aptamer is administered by intravenous administration, intrasynovial administration, transdermal administration, intramuscular administration, subcutaneous administration, or topical administration to a blood vessel.

26. The method of claim 25 , wherein the aptamer is administered by subcutaneous administration.

27. The method of claim 1 , wherein the host is suffering from cardiovascular disease, and wherein the cardiovascular disease is selected from the group consisting of: atheroscleosis, thrombophilia, embolisms, cardiac infarction, thromboses, angina, stroke, septic shock, hypertension, hyper-cholesterolemia.
