Abstract

A material includes a surface and a reactive agent that is located at the surface of the material, covalently attached to a backbone of the material, and/or located within the material. The reactive agent has nitrite reductase activity, nitrate reductase activity, and/or nitrosothiol reductase activity. The reactive agent also converts at least one of nitrites, nitrates and nitrosothiols to nitric oxide when in contact with blood. A reproducible nitrosothiol sensor is also disclosed.

Claims (24)

1. A method of increasing the biocompatibility of a medical device, the method comprising:

applying to a surface of the medical device a film including a reactive agent; and

contacting the medical device with blood, thereby reacting the reactive agent with at least one of nitrite, nitrate, or nitrosothiols endogenous in the blood to convert the at least one of nitrite, nitrate, or nitrosothiols to nitric oxide, and increase the biocompatibility of the medical device.

2. The method of claim 1 wherein the reactive agent includes a copper ion.

3. The method of claim 2 wherein the reactive agent further includes a neutral carrier ligand.

4. The method of claim 1 wherein the film further includes a polymer.

5. The method of claim 4 wherein the reactive agent is covalently bonded to the polymer.

6. The method of claim 5 wherein the reactive agent includes a metal ion ligand complex.

7. The method of claim 6 wherein the metal ion ligand complex includes a copper ion.

8. The method of claim 4 wherein the surface of the medical device to which the film is applied includes a polymer.

9. The method of claim 4 wherein the surface of the medical device to which the film is applied includes a metal.

10. The method of claim 4 wherein the polymer is selected from polyurethane, poly(vinyl) chloride, and polyhydroxyethylmethyloxylate.

11. The method of claim 4 wherein the polymer is selected from polymethyl methacrylate and silicone rubber.

12. The method of claim 1 wherein the step of applying is accomplished by dip-coating the medical device with a polymer including the reactive agent.

13. A method of increasing a time of useful contact of a medical device with blood, the method comprising:

coating the medical device with a composition including a reactive agent that reacts with at least one of nitrite, nitrate, or nitrosothiols endogenous within the blood to convert the at least one of nitrite, nitrate or nitrosothiols to nitric oxide; and

contacting the coated medical device with the blood to react the reactive agent with the at least one of nitrite, nitrate or nitrosothiols to generate the nitric oxide.

14. A method of increasing the biocompatibility of a medical device for use in vivo, the method comprising:

applying to a surface of the medical device a film including a reactive agent;

wherein:

the reactive agent generates nitric oxide when in contact with at least one of nitrite, nitrate, or nitrosothiols;

the generation of nitric oxide by the reactive agent increases the biocompatibility of the medical device;

the reactive agent includes a copper ion and a neutral carrier ligand; and

the neutral carrier ligand is an N x donor macrocyclic ligand and x is chosen from 2, 4, 5, 6 or 8.
