Abstract

A family of insecticidal polypeptides expressed in the venom gland of spiders of the genera Atrax and Hadronyche have been described. Also included are polynucleotides and expression vectors encoding the polypeptides and insect viruses and cells expressing the polypeptides. Transgenic plants and insects expressing the insecticidal polypeptides are also described. The insecticidal polypeptides may be employed in methods and compositions for treating insects, insect larvae, and plants.

Claims (16)

1. An isolated polynucleotide encoding: a polypeptide comprising an amino acid sequence having greater than or equal to about 90% identity to any one of SEQ ID NOs: 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27, wherein the polypeptide has insecticidal activity or a complement thereof.

2. The polynucleotide of claim 1 , encoding a polypeptide comprising any one SEQ ID NOs. 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27.

3. The isolated polynucleotide of claim 1 , wherein the polynucleotide further comprises polynucleotide sequences encoding a pro sequence, a signal peptide sequence, or a combination thereof, in frame with the encoded polypeptide.

4. The isolated polynucleotide of claim 1 , wherein the polynucleotide encodes a polypeptide comprising any one of SEQ ID NOs: 1, 5, 8, 11, 14, 17, 20, 23 or 26.

5. The isolated polynucleotide of claim 1 , wherein the polypeptide comprises an amino acid sequence having greater than or equal to about 90% identity to SEQ ID NO: 2, wherein the polypeptide has insecticidal activity.

6. An expression vector comprising a polynucleotide encoding any one of SEQ ID NOs. 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27 or a complement thereof, said polynucleotide operably linked to an expression control sequence.

7. The expression vector of claim 6 , wherein the expression vector is suitable for expression in an insect cell or a bacterial cell.

8. The expression vector of claim 6 , wherein the expression vector is a baculovirus vector.

9. A host cell comprising an expression vector comprising a polynucleotide encoding any one of SEQ ID NOs. 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27 or a complement thereof, said polynucleotide operably linked to an expression control sequence.

10. The host cell of claim 9 , wherein the host cell is a plant cell, a bacterial cell or an insect cell.

11. A transgenic insect virus comprising a polynucleotide encoding a polypeptide comprising an amino acid sequence having greater than or equal to about 90% identity to any one of SEQ ID NOs: 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27, wherein the polypeptide has insecticidal activity.

A transgenic plant, comprising a polynucleotide encoding a polypeptide which expresses a U-ACTX polypeptide, wherein the U-ACTX polypeptide comprises an amino acid sequence that is greater than or equal to about 90% identical to any one of SEQ ID NOs: 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27, wherein the U-ACTX polypeptide has insecticidal activity.

13. The transgenic plant of claim 12 , wherein the U-ACTX polypeptide comprises any one of SEQ ID NOs. 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27.

14. A transgenic insect comprising a polynucleotide encoding a polypeptide comprising an amino acid sequence having greater than or equal to about 90% identity to any one of SEQ ID NOs: 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27, wherein the polypeptide has insecticidal activity.

15. The transgenic insect of claim 14 , wherein the polypeptide comprises any one of SEQ ID NOs. 2, 3, 6, 9, 12, 15, 18, 21, 24 or 27.

16. The isolated polynucleotide of claim 1 , wherein the polynucleotide encodes an amino acid sequence having greater than or equal to about 95% identity to SEQ ID NO: 2.
