CGRP receptor antagonists
View Patent ↗The present invention is directed to compounds of Formula I: (where variables R 1 , R 2 , R 3 , R 4 , A, B, G, J, Q, T, U, V, W, X and Y are as defined herein) useful as antagonists of CGRP receptors and useful in the treatment or prevention of diseases in which the CGRP is involved, such as headache, migraine and cluster headache. The invention is also directed to pharmaceutical compositions comprising these compounds and the use of these compounds and compositions in the prevention or treatment of such diseases in which CGRP is involved.
1. The compound
or a pharmaceutically acceptable salt thereof.
2. A pharmaceutical composition comprising the compound of claim 1 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.
3. The pharmaceutical composition of claim 2 , which further comprises acetaminophen.
4. The pharmaceutical composition of claim 2 , which further comprises a triptan selected from the group consisting of sumatriptan, naratriptan, zolmitriptan, eletriptan, almotriptan, frovatriptan, donitriptan and rizatriptan.
5. The pharmaceutical composition of claim 4 , wherein the triptan is rizatriptan.
6. The pharmaceutical composition of claim 2 , which further comprises a non-steroidal anti-inflammatory agent or a cytokine-suppressing anti-inflammatory agent selected from the group consisting of aspirin, ibuprofen, ketoprofen, fenoprofen, naproxen, indomethacin, sulindac, meloxicam, piroxicam, tenoxicam, lornoxicam, ketorolac, etodolac, mefenamic acid, meclofenamic acid, flufenamic acid, tolfenamic acid, diclofenac, oxaprozin, apazone, nimesulide, nabumetone, tenidap, etanercept, tolmetin, phenylbutazone, oxyphenbutazone, diflunisal, salsalate, olsalazine and sulfasalazine.
7. The pharmaceutical composition of claim 6 , wherein the non-steroidal anti-inflammatory agent or cytokine-suppressing anti-inflammatory agent is selected from the group consisting of aspirin, ibuprofen and naproxen.
8. A method of treating headaches in a person in need of such treatment, comprising administering to the person a therapeutically effective amount of a compound of claim 1 or a pharmaceutically acceptable salt thereof.
9. The method of claim 8 , wherein the headaches are migraine headaches.
10. The method of claim 8 , wherein the headaches are cluster headaches.
