Abstract

A device is provided for chromatographic separations comprising a manifold comprising a plurality of connectors for connecting to one or more chromatographic separation columns and/or feed or extraction tubing. At least one central duct is formed between at least two connectors forming an inlet and an outlet respectively. The central duct comprises a closable duct valve; and a plurality of branch ducts branching from the central duct to a branch connector. The branch duct comprises a closable branch valve, wherein at least one branch duct is positioned between the inlet and the central duct valve and wherein at least one branch duct is positioned between the outlet and the central duct valve. The device is arranged for carrying out single-column and (continuous) multicolumn chromatographic separations. This allows the purification of biopharmaceutical products without having to develop, demonstrate and validate cleaning procedures for the valves.

Claims (21)

1. Device for chromatographic separations comprising:

a manifold comprising a plurality of connectors for connecting to one or more chromatographic separation columns and/or feed or extraction tubing;

at least one central duct between at least two connectors forming an inlet and an outlet respectively, the duct comprising a closable duct valve; and

a plurality of branch ducts branching from the central duct to a branch connector, the branch duct comprising a closable branch valve,

wherein at least one branch duct is positioned between the inlet and the central duct valve and wherein at least one branch duct is positioned between the outlet and the central duct valve wherein the device is formed as a connectable modular piece.

2. Device according to claim 1 , wherein at least one branch duct comprises another branch to form a branch duct between at least two branch connectors, the branch duct in fluid communication and closable from the central duct.

3. Device according to claim 2 , wherein the branch connectors are located opposite to each other.

4. Device according to claim 1 , wherein the device is formed to group said connectors and valves in a compact single piece manifold.

5. Device according to claim 2 , wherein said single piece manifold is formed as a disposable item.

6. Device according to claim 1 , wherein at least one of the duct valves and/or branch valves comprises a membrane valve, actuated by a separate actuator.

Device according to claim 1 , wherein the device is formed as a connectable modular piece comprising connectors to connect to a second connectable modular piece to form a plurality of parallel and or serially coupled central ducts so that at least one coupled branch duct that is connectable to a selectable plurality of central ducts via a selected number of branch valves in said modular pieces.

8. Device according to claim 7 , wherein the connectors are formed by corresponding gaskets provided on side walls of the modular piece to be pressed together by a press and providing a fluid communication between respective branch ducts and/or central ducts.

9. Device according to claim 7 , wherein the device further comprises a front member for providing system feed and extraction flows to the modular piece; and/or a rear member for providing a stop member for selectively closing ducts in the modular piece.

10. Device according to claim 1 , wherein the manifold is formed from a duct layout part and a membrane part for closing the duct layout part and for forming the membrane valves, and comprising mountings for mounting a corresponding number of actuators.

Device according to claim 10 , wherein the duct layout part comprises a central part coupled between a base part and a cover part; one side of the central part corresponding to a branch duct structure; and an opposite side of the central part corresponding to a central duct structure, the central part comprising through holes to correspond with through holes provided in a cover part; the cover part being coupled to the membrane part for closing a through hole so as to form a membrane valve between the central duct and the branch duct.

12. Device according to claim 10 , wherein the actuators are formed to correspond with membrane valves formed in the duct layout part.

13. Device according to claim 1 , wherein the connectors are formed of sanitary couplings.

14. Device according to claim 1 , wherein three extraction branches are coupled to the central duct between the inlet and the duct valve, and wherein two feeding branches are coupled to the central duct between the duct valve and the outlet.

15. Device according to claim 1 , wherein four to six extraction branches are coupled to the central duct between the inlet and the duct valve, and wherein six to eight feeding branches are coupled to the central duct between the duct valve and the outlet.

A system for providing chromatographic separations, comprising one or more devices according to claim 1 ; one or more chromatographic separation columns; said at least one or more devices connected to an inlet and/or outlet of at least one of said columns; feeding tubing and/or extraction tubing, at least some of the feeding and/or extraction tubing connected to branching ducts and a plurality of pumps to provide a flow in any of said columns and/or tubing.

17. A method for providing biopharmaceutical products involving a chromatographic separation process, the method comprising separating biopharmaceutical products with a system according to claim 16 .
