Abstract

Vectors and methods for the production of influenza viruses suitable as recombinant influenza vaccines in cell culture are provided. Bi-directional expression vectors for use in a multi-plasmid influenza virus expression system are provided. Additionally, the invention provides methods of producing influenza viruses with enhanced ability to replicate in embryonated chicken eggs and/or cells (e.g., Vero and/or MDCK) and further provides influenza viruses with enhanced replication characteristics.

Claims (9)

1. A reassortant influenza A virus comprising an altered H3 HA protein that is replication enhanced relative to the virus without the altered H3 HA protein, wherein the altered H3 HA protein comprises substitutions at positions 186 and 196; wherein the substitutions change the amino acids at positions 186 and 196 to valine and threonine, respectively.

2. The reassortant influenza virus of claim 1 , further comprising a substitution at position 226; wherein the substitution changes the amino acid at position 226 to isoleucine.

3. The reassortant influenza virus of claim 1 , wherein the influenza virus grows to a titer of at least 8.0 log 10 PFU/ml in embryonated eggs.

4. The reassortant influenza virus of claim 3 , wherein the influenza virus grows to a titer of at least 9.0 log 10 PFU/ml in embryonated eggs.

5. The reassortant influenza virus of claim 1 , wherein the influenza virus is cold adapted, temperature sensitive and attenuated.

6. The reassortant influenza virus of claim 1 , wherein the influenza virus comprises the backbone of the MDV-A strain or PR8 strain.

7. An immunogenic composition comprising the reassortant influenza virus of claim 1 .

8. An influenza vaccine comprising the reassortant influenza virus of claim 1 .

9. A live cold adapted, attenuated temperature sensitive influenza vaccine comprising the reassortant influenza virus of claim 1 .
